Motolice nad eledi Paramphistomoidea p edstavují významné patogeny napadající r zné druhy obratlovc v etn hospodá sky významných p ežvýkavc a prasat a zp sobují závažné ekonomické škody. Dosp lí jedinci se nachází v p edžaludcích, p ípadn žlu ovodech i tlustém st ev hostitele. Vážné patologické zm ny až smrt hostitele však zp sobují p edevším juvenilní stádia v prepatentní period lokalizovaná v tenkém st ev . Diagnostika nákaz paramfistomními motolicemi je provád na p edevším koprologickými metodami (McMasterova metoda, mini-FLOTAC, sedimenta ní metody), které detekují p ítomnost vají ek v trusu hostitele. B hem prepatentní periody však nemohou být tyto metody použity. Proto jsou vyvíjeny metody imunologické, které mohou zachytit nákazu i v její rané fázi. Sou asn je kladen d raz i na vývoj metod pro molekulární identifikaci druhu, které by umožnily provád t epidemiologické studie a zhodnotit ekonomický význam jednotlivých druh paramfistomních motolic. Tato práce shrnuje poznatky o sou asných možnostech diagnostiky paramfistomózy a o vývoji p ístup , které by se v budoucnosti mohly uplatnit v diagnostice nebo p i identifikaci druh paramfistomních motolic. Trematodes of the superfamily Paramphistomoidea are significant pathogens of various species of vertebrates including livestock. They also cause significant economic losses. Adults are located in the forestomaches, eventually in the bile ducts or in the large intestine of the host. Severe pathological changes and sometimes even death of the host are caused by juvenile stages during their migration in small intestine. The diagnosis of paramphistomosis is done mainly by coprological methods (McMaster method, mini-FLOTAC, sedimentation methods) which detect the presence of eggs in the faeces of definitive host. However, these methods cannot be used during the prepatent period. Therefore it is important to develop immunological methods that enable detection of the infection in the early phase. It is also important to develope methods for molecular identification of paramphistomes to facilitate epidemiological studies and evaluate the economic importance of individual species of Paramphistomoidea. Presented thesis summarizes current methods and development of new strategies in diagnostics of paramphistomosis and molecular identification of paramphistomes.
