Jednobuněčné organismy jako jsou kvasinky, byly dříve převážně studovány v třepaných kulturách a nikoli na pevném povrchu, jak je tomu běžné v přírodě. V přírodě totiž skoro žádná buňka nežije osamoceně, ale naopak často tvoří mnohobuněčné kolonie nebo biofilmy. V současné době již řada studií probíhá i na pevném médiu, tedy za podmínek podobnějších podmínkám přírodním. Naše laboratoř vyvinula speciální techniky pro výzkum kolonií kvasinky Saccharomyces cerevisiae. Tyto techniky nám umožňují popis a zkoumání jednotlivých subpopulací a buněk v kvasinkové kolonii. Cílem této práce bylo připravit řadu mutantních kmenů produkujících cílové geny mitochondriální RTG signalizace, popsat morfologii a ultrastrukturu jejich kolonií a pomocí nich přispět k objasnění funkce retrográdní dráhy při vývoji kvasinkové kolonie. Práce popisuje expresi několika vybraných genů (CIT2, RTG1, RTG2, RTG3) ve výchozím kmeni BY4742 a v dalších mutantních kmenech s delecí jednoho či více regulačních genů RTG dráhy. Výsledky získané v rámci diplomové práce spolu s výsledky dalších autorů se staly součástí vědecké publikace (Podholová et al., 2016). Powered by TCPDF (www.tcpdf.org) Unicellular organisms such as yeast have been traditionally studied in shaken cultures, i.e., under condition in which they do not grow attached to solid surfaces as under natural conditions. In nature, cells only rarely live alone, but, on the other hand often create multicellular colonies or biofilms. During last years, yeasts started to be investigated also when grown on solid media. Our laboratory has previously developed special techniques for investigation of yeast colonies. These techniques allowed us to describe individual cell subpopulations within the colonies. The aim of this work was to prepare a series of mutant strains, describe morphology and ultrastructure of their colonies with the aim to contribute to understanding ofthe role of mitochondrial retrograde signalling pathway in the development of yeast colonies. This work describes expression of few selected genes (CIT2, RTG1, RTG2, and RTG3) in colonies of the parental strain BY4742 and of other mutant strains with deletion of one or more genes of RTG regulatory pathways.
