Hlavním cílem této práce byla detailní charakterizace interakce DNA-vazebné domény proteinu FOXO4 se dvěmi kanonickými sekvencemi DNA a dále objasnění úlohy proteinu 14-3-3 v procesu regulace funkce FOXO4. Transkripční faktory FOXO jsou účinnými aktivátory transkripce genů, čímž ovlivňují celou řadu buněčných procesů. Protein FOXO4 patří do rodiny forkhead transkripčních faktorů, což je skupina několika desítek proteinů, jejichž společným znakem je velmi konzervovaná DNA-vazebná doména. Podstata DNA-vazebné specifity těchto proteinů, tedy co přesně determinuje drobné odlišnosti ve vazebných vlastnostech jednotlivých forkhead proteinů, je i přes množství dostupných strukturních dat stále nejasné. Proto byla provedena detailní charakterizace interakcí mezi DNA-vazebnou doménou proteinu FOXO4 a DNA pomocí rezonance povrchového plazmonu (SPR) a časově- rozlišené fluorescenční spektroskopie. The main aim of this work was detailed characterization of interaction between DNA binding domain of protein FOXO4 with two canonical DNA sequences and further clarifying the role of the 14-3-3 protein in the regulation of activity of protein FOXO4. FOXO transcription factors are potent activators of the transcription of genes, which affect a variety of cellular processes. FOXO4 protein belongs to the family of forkhead transcription factor, which is a group of several tens of proteins, whose common feature is a highly conserved DNA- binding domain.
