Glutamátový receptor δ 2 (GluRδ2) je exprimován v dendritech Purkyňových buněk lokalizovaných v kůře mozečku. Správná funkce GluRδ2 je nezbytná pro zdravý vývoj mozečku, formování synapsí mezi paralelními vlákny granulárních buněk a Purkyňovými buňkami a také pro vznik dlouhodobé deprese důležité pro utváření paměti. Lurcher mutace, lokalizovaná na 6. autozomálním chromosomu, mění tento receptor substitucí aminokyseliny ve třetí transmembránové doméně v konstitutivně otevřený iontový kanál. Následkem této změny dochází k úplnému vymření populace Purkyňových buněk a silné degeneraci granulárních buněk a buněk oliverních neuronů. Myši postižené Lurcher mutací mají sníženou hmotnost a samice rodí menší počty mláďat. Fertilita samců není mutací ovlivněna. Po behaviorální stránce vykazují Lurcher mutanti značné deficity. Trpí ataxií, typickou pro všechny cerebelární neurodegenerace, mají sníženou schopnost ve fyzických výkonech, zhoršenou schopnost orientace a nejsou téměř schopni učení. Tato práce shrnuje současné poznatky o Lurcher mutaci od její molekulární podstaty po behaviorální projevy. Specifické znaky této degenerace nám dovolují zkoumat vliv neurodegenerativních poruch mozečku na celkové kognitivní funkce mozku, objevovat jejich příčiny a nalézat možnosti léčby. Lurcher mutanti jsou vhodnými... The glutamate receptor δ 2 (GluRδ2) is expressed in dendrites of Purkinje cells localized in the cerebellar cortex. Correct function of GluRδ2 is necessary for cerebellar development, synapse formation between parallel fibers of the granular and Purkinje cells and for inducing long term depression important in memory formation. Lurcher mutation, localized to 6th autosomal chromosome, transforms GluRδ2 into constitutively open ion channel by amino acid substitution in third transmembrane domain. As a result, almost complete disappearance of Purkinje cells population and a large degeneration of granular cells and olivary neurons occurs. Mice impaired by Lurcher mutation have lower body weight and reduced litter size. Fertility of males is not affected. Lurcher mutants display extensive behavioral deficits. Mice suffer from ataxia typical for cerebellar neurodegenerations. They have reduced physical performance, impaired spatial orientation and learning capabilities. The aim of this work is to summarize recent knowledge about Lurcher mutation from molecular basis to behavioral manifestation.
