[cze][eng] Mechanismy translokace přes plazmatickou membránu eukaryotických buněk jsou důležitým faktorem virulence patogenních bakterií způsobujících onemocnění eukaryotickým organismům. Toxiny translokují své vlastní domény s toxickou aktivitou dovnitř buňky nebo vytvoří v její membráně pór, kterým mohou procházet další molekuly od iontů až po DNA, RNA nebo proteiny. Výzkum translokace těchto látek umožňuje charakterizovat mechanismus translokace a také i vlastnosti pórotvorných toxinů. Několik těchto toxinů je natolik podrobně popsáno, že po cílené mutagenezi se dají využívat pro určování vlastností molekul, které jejich póry prochází. Jednou z takových aplikací je cílená doprava léčiv do rakovinných buněk pomocí Shiga toxinu nebo měření průchodu nukleotidů i celých vláken nukleových kyselin přes membránový kanál vytvářený α-hemolysinem S. aureus. Tato metoda si v současné době našla uplatnění při sekvenaci DNA. Klíčová slova: translokace, bakteriální toxiny, plasmatická membrána, nanopórové sekvenováníThe bacterial protein toxins endowed with the ability to translocate across the plasmatic membrane are often crucial virulence factors of pathogenic bacteria invading eukaryotic organisms. These toxins translocate either their own protein domains carrying toxic activity or can form pores transferring other substances like small ions, DNA, RNA or proteins. By observing the translocation of these molecules together with other artificially prepared agents on synthetic membranes it allows detailed understanding of mode of action of individual pore-forming toxins. Some of the toxins were actually described in such a detail that can serve as tools for drug delivery or characterization of new translocated molecules. One of such examples is the transfer of nucleotides or the whole nucleic acid molecules across the membrane pore of S. aureus α-hemolysine.
