[cze][eng] Diplomová práce se zabývá teoretickými i praktickými aspekty vysokoúčinné kapalinové chromatografie (HPLC). Představuje ji jako jednu z nejčastěji používaných analytických metod. V teoretické části pojednává o instrumentaci a jednotlivých součástech sestavy pro HPLC. Věnuje se také stručnému popisu nejběžnějších mobilních a stacionárních fází, používaných v tomto analytickém postupu. Na základě teoretických poznatků je připravena laboratorní úloha pro vysokoškolské studenty, budoucí učitele chemie. Jako vhodná látka pro tuto učební úlohu byl vybrán kofein. Jedná se o látku nejen všeobecně známou, ale také hojně využívanou například v potravinářství. Součástí práce je stručný pohled na rámcové vzdělávací programy (RVP) z hlediska možného zařazení HPLC do výuky v primárním a sekundárním vzdělávání. Přílohou k práci je návod k sestavené laboratorní úloze. This method is introduced as one of the most frequently used current analytical methods. The theoretical part of thesis is focused on instrumentation of HPLC and particular components of HPLC analytical system. The most often used mobile phases and static phases are described as well. Based on these theoretical aspects, laboratory exercise using HPLC for future teachers is designed. Caffeine is used as ideal model material for this exercise. Caffeine is well-known substance, because of its traditional usage for example in food-processing industry. Final part of this thesis is brief view on framework educational programmes for primary and secondary education. As appendix of this thesis, manual for referenced laboratory exercise is provided.
