Jandovská, Kateřina: Příprava analogů ceramidů a hodnocení jejich vlivu na bariérovou funkci kůže. Ceramidy patří mezi sfingolipidy, jejichž molekulu tvoří sfingoidní báze a dlouhá mastná kyselina. Jsou dosud známy nejenom jako důležití druzí poslové hrající významnou úlohu v buněčné diferenciaci, proliferaci a apoptóze, ale zároveň i jako podstatná složka funkční kožní bariéry. Ačkoli jsou tyto molekuly intenzivně studovány, přesný vliv jejich struktury na bariérovou funkci kůže není znám. Cílem mé práce bylo studovat vliv délky acylu a stereochemie na C3 u dihydroceramidů (tedy ceramidů bez dvojné vazby v poloze 4) na prostupnost modelových membrán simulujících kožní bariéru. Mým úkolem byla syntéza tří ceramidů s krátkým acylovým řetězcem o čtyřech uhlících (odvozených od dihydrosfingosinu (dS), L-threo-dihydrosfingosinu (L-dS)‚ a L-threo-sfingosinu (L-S)), příprava modelových membrán stratum corneum (SC) obsahujících dihydroceramidy s délkami acylu C2, C4, C6, C8 a C24 a připravené stereoizomery C4-ceramidů a C4-dihydroceramidů a následné hodnocení jejich elektrické impedance a permeability pro dvě modelová léčiva. Vliv připravených produktů nanesených na modelové membrány na permeabilitu těchto membrán byl hodnocen ve Franzových difuzních celách pomocí měření elektrické impedance a dvou... Ceramides belong to sphingolipids, their molecule is formed by a sphingoid base and long fatty acid. They are known not just as important second messengers playing a significant role in cell differentiation, proliferation and apoptosis, but also as essential part of functional skin barrier. Although these molecules are studied intensively, the exact effect of their structure on barrier function of the skin is poorly understood. The aim of my work was to study the effect of acyl chain length and stereochemistry on C3 of dihydroceramides (ceramides with single bond on C4) on the permeability of model membranes simulating the skin barrier. I have synthetized 3 ceramides with short acyl chain of 4 carbons (derived from dihydrosphingosine (dS), L-threo-dihydrosphingosine (L-dS) and L-threo-sphingosine (L-S)), and prepared model membranes of stratum corneum (SC) containing dihydroceramides with C2, C4, C6, C8 and C24 acyl chain length and stereoisomeres of C4-ceramides and C4-dihydroceramides as well.
