Metoda real-time PCR je varianta klasické PCR, která umožňuje kontinuálně monitorovat přírůstky DNA během každého cyklu a využívá se především k analýze genové exprese. Na základě předchozích experimentů jsme se rozhodli otestovat účinek antikoagulačních činidel EDTA, heparin, citrát sodný a CPDA na expresi vybraných genů imunologického spektra, a dále doby času od odběru krve do zpracování vzorku na hladiny mRNA vybraných genů, jež jsou dány změnami genové exprese a/nebo degradací mRNA. Pro kvantifikaci mRNA sledovaných genů jsme z leukocytů periferní krve izolovali celkovou RNA a tu přepsali pomocí reverzně transkriptázové PCR do cDNA, jež sloužila jako templát pro real-time PCR. Pro sledování změn exprese způsobených vlivem jednotlivých antikoagulačních činidel byla použita krev od 10 dobrovolníků (každý dárce krve byl odebrán do 3 vakuových zkumavek s antikoagulačním činidlem EDTA, heparinem a citrátem sodným). Deset vzorků buffy coatů jsme získali od dárců krve v transfuzních vacích s antikoagulačním agens CPDA. Buňky krve ovlivněné CPDA vykazovaly statisticky významně nižší hladiny mRNA genů TLR4, MYC a CCL2 ve srovnání s buňkami krve odebrané do jednoho ze 3 typů vakuet, a naopak, hladiny mRNA IL-10 a TNFa byly v buňkách těchto vzorků signifikantně zvýšeny. Antikoagulační agens používaná v... Real-time PCR method is a type of PCR which allows continual monitoring of DNA amplification during every cycle of its process. It is mostly used for gene expression analysis. Based on the results of previous experiments, we decided to test out the effect of anticoagulants EDTA, heparin, sodium citrate and CPDA on the expression of selected genes of the immunological spectrum and further, to test how the time period between drawing the blood and processing of blood sample influences mRNA levels of selected genes that are determined by changes in gene expression and/or mRNA degradation. To quantify mRNA of the studied genes, we isolated total RNA from the peripheral blood leucocytes and transcribed it into cDNA by using the reverse transcription PCR. This cDNA served as a template for the real-time PCR. To examine the changes of the expression caused by the effect of each particular anticoagulants, peripheral blood derived from 10 volunteers was used (each donor's blood was taken into 3 vacuum tubes with EDTA, heparin and sodium citrate anticoagulant agents). Next to that, we obtained 10 buffy coat samples in transfusion blood bags with CPDA anticoagulant agent.
