Aristolochové kyseliny (AA) jsou lidskými karcinogeny, které rovněž vykazují silné nefrotoxické vlastnosti. Směs AA je obsažena v rostlinách čeledi podražcovitých (Aristolochiaceae), které byly a nadále jsou užívány v tradiční medicíně, zejména v Asii. Aristolochové kyseliny se podílí na vzniku dvou typů nefropatií. Jedna z nich dokonce nese označení nefropatie vyvolaná aristolochovou kyselinou (Aristolochic Acid Nephropathy, AAN). Druhým onemocněním je balkánská endemická nefropatie (Balkan Endemic Nepropathy, BEN). Obě onemocnění jsou doprovázena malignacemi v urotheliální tkáni, které jsou vyvolány karcinogenním působením AA. Zvláštním znakem AAN a BEN je skutečnost, že ne všichni pacienti vystavení působení AA byli postiženi nefropatií a vývojem nádorového onemocnění. Jednou z příčin této skutečnosti mohou být rozdíly v aktivitách a expresních hladinách enzymů biotransformujících AAI, hlavní toxickou složku obsaženou v rostlinném extraktu. Detailní poznání enzymů podílejících se na metabolismu AAI může přispět k objasnění interindividuální vnímavosti vůči AAN, BEN a malignacím. Aristolochová kyselina I je v organismu buď oxidačně detoxifikována anebo redukčně aktivována. Redukční bioaktivace AAI vede ke tvorbě kovalentních aduktů AA s DNA, které resultují ve vznik unikátní transversní mutace... Aristolochic acids (AA) are human carcinogens which have also very strong nephrotoxic properties. A mixture of AA is present in Aristolochiacae plant species. These plants were and still are used in traditional medicine in some countries, particularly in Asia. Aristolochic acids participate in development of two types of nephropathies. The first disease is designated as Aristolochic Acid Nephropathy (AAN), the second one is Balkan Endemic Nephropathy (BEN). Both nephropathies are associated with urothelial malignancies, which are caused by AA. One of the common features of ANN and BEN is that not all individuals exposed to AA suffer from nephropathy and tumour development. One cause for these different responses may be individual differences in the activities and expression levels of the enzymes catalyzing the biotransformation of AAI, the major toxic component of AA contained in Aristolochia species. Detailed knowledge of enzymes which participate in metabolism of AAI may contribute to elucidation of inter-individual susceptibility to AAN, BEN and later urothelial malignancies. Aristolochic acid I is either oxidative detoxicated or reductive activated by biotransformation enzymes.
