Modulace nádorového mikroprostředí představuje možnost, jak inhibovat růst nádorů a posílit protinádorovou imunitní odpověď. V předložené práci jsme použili pro modulaci nádorového mikroprostředí dvě strategie. Zaprvé, jsme zkonstruovali rVACV ko-exprimující tumor supresorový gen pro vazebný protein typu 3 pro růstové faktory podobné inzulinu (IGFBP-3) a gen kódující fúzní imunogen SigE7LAMP. Exprese IGFBP-3 byla řízena buď časným vakciniovým promotorem H5 nebo syntetickým časně/pozdním (E/L) promotorem. Zjistili jsme, že exprese IGFBP-3 řízená H5 promotorem vede k produkci většího množství proteinu než exprese řízená E/L promotorem. Imunizace myší rVACV P13-SigE7LAMP-H5- IGFBP-3 inhibovala růst nádorů TC-1 efektivněji než imunizace kontrolním virem P13-SigE7LAMP-TK- . Zároveň indukovala také silnější odpověď T-lymfocytů proti VACV antigenům. Pozorovali jsme, že vysoká exprese IGFBP-3 vedla ke zvýšení míry replikace viru a to jak in vitro, tak in vivo, což mělo za následek déletrvající antigenní stimulaci. Vysoká míra produkce IGFBP-3 rovněž korelovala s lepší adsorpcí viru P13-SigE7LAMP-H5-IGFBP-3 na buňky CV-1 v porovnání s P13-SigE7LAMP-TK- . Identifikovali jsme dva rozdíly ve struktuře IMV mezi viry P13-SigE7LAMP-H5-IGFBP-3 a P13-SigE7LAMP-TK- . IMV P13-SigE7LAMP-H5-IGFBP-3 do své struktury... Modulation of the tumor microenvironment represents a possible way to inhibit cancer growth and enhance anti-cancer immune responses. In the presented work we employ two strategies for tumor microenvironment modulation. Firstly, we have constructed rVACV co-expressing the tumor suppressor gene insulin-like growth factor-binding protein-3 (IGFBP- 3) and the fusion gene encoding the immunogen SigE7LAMP. The expression of IGFBP-3 was regulated either by the early vaccinia virus H5 promoter or by the synthetic early/late (E/L) promoter. We have shown that expression of IGFBP-3 regulated by the H5 promoter yielded higher amounts of IGFBP-3 protein when compared with the E/L promoter. Immunization with P13-SigE7LAMP-H5-IGFBP-3 was more effective in inhibiting the growth of TC-1 tumors in mice and elicited a higher T-cell response against VACV-encoded antigens than the control virus P13-SigE7LAMP-TK- . We found that high-level production of IGFBP-3 enhanced virus replication both in vitro and in vivo, resulting in profound antigen stimulation. Production of IGFBP-3 was associated with a higher adsorption rate of P13-SigE7LAMP-H5-IGFBP-3 to CV-1 cells when compared with P13-SigE7LAMP-TK- .
