Teoretická část práce podává informace o biofarmaceutickém klasifikačním systému léčiv a jeho kontextu ve výzkumu a vývoji léčiv. Jsou prezentovány metody používané ke zvýšení a zrychlení rozpustnosti včetně elektrospiningu. Experimentální část je pilotní studií zaměřenou na hodnocení podmínek vhodných pro rozpouštění a testování nově připravených nanovlákených membrán z polyvinylpyrrolidonu s vysokým obsahem (do cca 35 procent) gatrinu jako látky špatně rozpustné ve vodných vehikulech. Parametry HPLC stanovení gatrinu za použití C18 sorbentu a mobilní fáze acetonitril: fosfátový pufr pH 8 byly předběžně vyhodnoceny jako bezproblémově použitelné pro vehikula typu fosfátového pufru o pH 6,0. Stejné podmínky se ukázaly být ve vážné kolizi s polymerním materiálem nanovlákenné membrány v průběhu disolučního hodnocení nebo s acetonitrilem v mobilní fázi, přesného stanovení gatrinu nebylo v tomto případě dosaženo. Tyto závěry vedou k návrhu na změnu formulace nanovlákenných membrán použitím polymeru odlišného od polyvinylpyrrolidonu (např. hydroxypropylcelulosy) nebo na záměnu acetonitrilu methanolem v mobilní fázi pro HPLC. Ve všech případech však bylo všechno analyticky dostupné gatrinu vždy rozpuštěno a stanoveno do cca 10 až 12 minut od startu rozpouštěcích testů, což lze považovat za vhodné jak pro... The theoretical part of thesis provides information on the biopharmaceutical classification system of drugs and its context in the research and development of pharmaceuticals. The methods used to increase the solubility and acceleration including electrospinning are presented. The experimental part is the pilot study on the evaluation of conditions suitable for dissolution testing of newly prepared nanofibers made from polyvinylpyrrolidone membranes with a high content (up to 35 per cent) gatrin as a substance poorly soluble in aqueous vehicles. The parameters of determination of gatrin by HPLC using C18 sorbent and a mobile phase of acetonitrile: phosphate buffer pH 8 were preliminarily evaluated as perfectly applicable to vehicle type-phosphate buffer pH 6.0. The same conditions were found to be in a severe collision with a polymer material of nanofibrous membrane during the dissolution evaluation or with acetonitrile in the mobile phase, an accurate determination of gatrin was not obtained in this case. These findings lead to the proposal to change the formulation of the nanofiber membranes using polymer different from polyvinylpyrrolidone (eg. hydroxypropylcellulose) or the replacement of acetonitrile for methanol at the mobile phase for HPLC.
