Tato bakalářská práce je zaměřena na separaci čtyř modelových proteinů - konalbuminu, hovězího sérového albuminu a beta-laktoglobulinů A a B - metodou gradientové chromatofokusace v uspořádání vysokoúčinné kapalinové chromatografie na dvou komerčních aniontově-výměnných kolonách. Jedna kolona obsahovala jako stacionární fázi silný anex (Agilent Bio SAX) a druhá slabý anex (Agilent Bio WAX). Pro separaci proteinů gradientovou chromatofokusací byly optimalizovány celkem tři různé pH gradienty mobilní fáze, jeden šedesátiminutový a dva třicetiminutové. Při použití všech tří testovaných gradientů došlo k separaci směsi konalbuminu s beta-laktoglobuliny A a B na základní linii na silném i slabém anexu při průtoku mobilní fáze 1 ml/min. Ve všech testovaných systémech se silným anexem jako stacionární fází bylo dosaženo separace všech tří izoforem hovězího sérového albuminu. Nejvyšších hodnot rozlišení píků při separaci směsi konalbuminu a beta-laktoglobulinů A a B bylo dosaženo za třicet minut na koloně obsahující silný anex jako stacionární fázi. Nejvyšších hodnot rozlišení píků izoforem hovězího sérového albuminu bylo dosaženo za šedesát minut na koloně obsahující silný anex jako stacionární fázi. Klíčová slova: gradientová chromatofokusace, separace proteinů, vysokoúčinná kapalinová chromatografie This bachelor thesis is focused on separation of four model proteins - conalbumin, bovine serum albumin, beta-lactoglobulin A and beta-lactoglobulin B - by gradient chromatofocusing high-performance liquid chromatography with two commercial anion-exchange columns. The first column contained a strong anion exchanger (Agilent Bio SAX) and the second one contained a weak anion exchanger (Agilent Bio WAX). Three different mobile phase pH gradients were optimized, one-hour gradient and two half-an-hour gradients. The baseline separation of mixture containing conalbumin and beta-lactoglobulines A and B was achieved on both anion- exchange columns using each of the three optimized gradients and with flow rate 1 ml/min. Three bovine serum albumin isoforms were resolved in all tested systems using a strong anion exchanger as the stationary phase. The highest resolution of mixture containing conalbumin and beta-lactoglobulines A and B was obtained with a strong anion exchanger as the stationary phase in thirty minutes. The highest resolution of bovine serum albumin isoforms was achieved with a strong anion exchanger as the stationary phase in sixty minutes.
