Tato bakalářská práce je zaměřena na studium retenčního chování tryptofanu a jeho derivátů a nalezení vhodných podmínek pro chirální separace těchto látek metodou vysokoúčinné kapalinové chromatografie. Sada chirálních analytů obsahovala D,L-tryptofan a 8 jeho derivátů - 5-fluoro-D,L-tryptofan, 5-hydroxy-D,L-tryptofan, N-BOC-D,L-tryptofan, D,L-tryptofanol, D,L-tryptofan metyl ester, D,L-tryptofan butyl ester, D,L-tryptofan oktyl ester a D,L-tryptofan benzyl ester. V této práci byly použity čtyři chirální stacionární fáze, a to na bázi cyklofruktanů (kolona Larihc CF6-P), makrocyklických antibiotik (kolony Chirobiotic V a T) a amylosy (kolona Chiralpak AD-RH). Separace byly prováděny ve dvou separačních módech - reverzním a polárně organickém. Nejlepší enantioseparace 5-fluoro-D,L-tryptofanu, 5-hydroxy-D,L-tryptofanu a D,L-tryptofanu bylo dosaženo na teikoplaninové chirální stacionární fázi. Všechny tyto tři analyty byly separovány na základní linii při použití polárně organického módu. Chirální stacionární fáze na bázi amylosy byla nejvhodnější pro enantioseparaci esterů tryptofanu. Tři ze čtyř esterů tryptofanu (D,L-tryptofan metyl ester, D,L-tryptofan butyl ester a D,L-tryptofan benzyl ester) byly částečně separovány na této chirální stacionární fázi v reverzním módu. Klíčová slova: HPLC,... This bachelor thesis is focused on study of retention and enantioseparation of tryptophan and its derivatives by high performance liquid chromatography. The set of chiral analytes contained D,L-tryptophan, 5-hydroxy-D,L-tryptophan, 5-fluoro- D,L-tryptophan, D,L-tryptophanol, D,L-tryptophan methyl ester, D,L-tryptophan butyl ester, D,L-tryptophan octyl ester, D,L-tryptophan benzyl ester and N-BOC-D,L-tryptophan. Four different chiral stationary phases were used, i.e. cyclofructan-based (Larihc CF6-P column), two macrocyclic antibiotics (Chirobiotic V and T columns) and amylose-based (Chiralpak AD-RH column). Reversed phase and polar organic separation modes were used. Enantiomers of 5-fluoro-D,L-tryptophan, 5-hydroxy-D,L-tryptophan and D,L-tryptophan were successfully separated on teicoplanin chiral stationary phase in polar organic mode. The amylose-based chiral stationary phase proved to be the most suitable for enantioseparation of esters of tryptophan. Three of them (D,L-tryptophan methyl ester, D,L-tryptophan butyl ester and D,L-tryptophan benzyl ester) were partially enantioseparated in reversed phase mode.
