[cze][eng] Na onkolytických vírusoch prebiehajú štúdie za účelom ich možného využitia ako protinádorových terapeutík. Očakáva sa, že napomôžu nielen priamym onkolytickým účinkom, ale v organizme nabudia i protinádorovú imunitu. V predkladanej práci sme skúmali dve HPV-16transformované myšacie bunkové línie, TC-1 a MK16, a reovírus typ 3, kmeň Dearing (RV). Obidve bunkové línie sú vysoko citlivé na účinky RV a in vitro produkujú značné množstvá infekčných viriónov. Napriek tomu sa od seba líšia. Keď sme naočkovali vysoké dávky infikovaných buniek do imunosuprimovaných zvierat, ich onkogénna aktivita bola silne potlačená, takmer úplne v prípade MK16, avšak menej u onkogénnejšej TC-1 línie. Keď boli imunizované zvieratá čelenžované TC-1 bunkami, ožiarené bunky boli lepším imunogénom než infikované bunky. Prekvapivo, keď boli čelenžované MK16 bunkami, opak bol pravdou. V ďalšej sérii experimentov sme demonštrovali, že RV sa prednostne replikuje v nádorových bunkách a že je schopný prekonať adaptáciu buniek na hypoxiu (1% O2), infikuje a taktiež zabíja hypoxické nádorové bunky. RV sa môže replikovať v hypoxickom nádorovom mikroprostredí i spôsobiť cytopatický efekt s následnou indukciou bunkovej smrti. Zistili sme, že veľké percento buniek v hypoxii je zabitých na kaspázach nezávislými mechanizmami. Navyše sme ukázali, že...Oncolytic viruses are examined to serve as anticancer therapeutics. It is expected that in addition to direct oncolytic effect their action will also help eliciting a solid antitumor immunity. In presented series of experiments we have employed two HPV16- transformed mouse cell lines, TC-1 and MK16, and reovirus type 3, strain Dearing (RV). Both cell lines are highly susceptible to RV and produce large amounts of infectious virus in vitro. Still, some differences were encountered. When inoculating high doses of infected cells into syngeneic animals their oncogenic activity was strongly suppressed, nearly completely in the case of MK16 cells but less efficiently in the case of more oncogenic TC-1 cells. When immunized animals were challenged with TC-1 cells, the irradiated cells proved to be a much better immunogen that the infected cells. However, when challenged with MK16 cells the opposite was true. In another study we demonstrate that RV replicates preferentially in tumor cells and that the virus is able to overcome cellular adaptation to hypoxia (1% O2) and infect and kill hypoxic tumor cells. RV can both replicate in a hypoxic tumor microenvironment and can cause cytophatic effect and subsequently induce cell death.
