Interakce proteinů s vazebnými partnery se vyskytují u všech živých organismů, takřka u každého buněčného procesu. Studium proteinových interakcí proto tvoří významnou část biochemického výzkumu. Ukazuje se, že ještě cennější, než získání rovnovážných konstant těchto dějů, je určení hodnot jednotlivých energetických složek, tedy změn enthalpie a entropie. Pro termodynamickou analýzu vzniku komplexu vazebných partnerů, tedy pro určení hodnot změn entropie a enthalpie, lze využít isothermální titrační kalorimetrii (ITC). Mikrokalorimetrie se tak stává důležitou optimalizační technikou při vývoji nových léčiv, například protivirových látek. Přestože je virus HIV známý přes 30 let, intenzivní výzkum nepřinesl ani vakcínu ani lék, který by trvale vyléčil pacienty. Bylo schváleno již 26 léků, z nichž většina cílí virové enzymy reversní transkriptasu a proteasu. Antiretrovirální léčba brání množení HIV a udržuje v chodu imunitní systém, avšak dlouhodobé užívání vede k resistenci vůči farmakům, která je způsobena mutacemi v cílových proteinech. Jeden z poměrně nových cílů terapeutického zásahu je formování kapsidy při vzniku nových virionů. Při skládání kapsidy dochází k mnoha protein - proteinovým interakcím, které jsou nezbytné při tvorbě infekčních virových částic. Jako účinný inhibitor skládání kapsidy... The interactions of proteins with their binding partners occur in every living organisms, in almost every cell process. Therefore the exploration of protein interactions forms significant part of biochemical research. It appears that even more valuable information than the value of equilibrium constants of these interactions is determination of individual energy components - changes in enthalpy and entropy. Thermodynamic analysis by isothermal titration calorimetry (ITC) can determine the changes in entropy and enthalpy caused by formation of complex of binding partners. Microcalorimetry is also an important optimization technique in development of new drugs, for example antiretrovirotics. Despite HIV is a virus known for over 30 years, intensive research has neither brought vaccine nor drug that would permanently cure patients. Already 26 drugs were approved, most of them target viral enzymes reverse transcriptase and protease. Antiretroviral treatment prevents the propagation of HIV and maintains immune system, but long - term use leads to resistance against drugs, which is caused by mutations in the target proteins.
