[cze][eng] Regulace genu DLX1, jehož signifikantně zvýšená hladina byla detekována právě u pacientů s akutní myeloidní leukémií s mutací FLT3-ITD, je dosud zcela neprozkoumaným tématem. Prvním cílem této práce bylo zjistit, které signální dráhy regulují expresi genu DLX1. Z těchto drah byly pomocí metod qRT-PCR a western blot vybrány dráhy ERK a JNK. Uvedené dráhy způsobují aktivaci podjednotek transkripčního faktoru AP-1, jehož předpokládané vazebné místo bylo identifikováno také na promotoru genu DLX1. Druhým cílem této práce bylo proto ověření hypotézy o předpokládané regulaci exprese genu DLX1 (přes ERK/JNK dráhu) probíhající přes AP-1 vazebné místo na promotoru. K ověření hypotézy byla použita metoda duální luciferázové eseje, která využívá luminiscenční aktivity luciferázy, jejíž gen je obsažen v použitém luciferázovém vektoru. Před tento gen byla při tvorbě konstruktů využívaných ve výše zmíněné eseji vložena nejprve krátká část promotoru genu DLX1 a poté i jeho dlouhá část (v okolí AP-1 místa). Z výsledků studie vyplývá, že regulace exprese genu DLX1 přes AP-1 vazebné promotorové místo je důležitou, nikoli však postačující součástí regulační kaskády probíhající přes ERK a JNK dráhu. Na regulaci exprese genu DLX1 se pravděpodobně podílejí rovněž jiné transkripční faktory, které jsou aktivovány ERK1/2 kinázou....Regulation of expression DLX1 gene, whose elevated levels are detected in patients with acute myeloid leukemia with FLT3-ITD mutations, is not still completely explored topic. The first aim of this study was to determine which selected signaling pathways regulate gene expression of DLX1. ERK a JNK pathways were selected by using qRT-PCR and western blot. These pathways cause activation of the transcription factor AP-1 subunits, the AP-1 putative promoter binding site was identified also in the promoter of the DLX1 gene. The second aim of this study was to test the hypothesis on the regulation of gene expression of DLX1 (via ERK/JNK pathway) through AP-1 binding site on the promoter. Dual luciferase assay using luminescent luciferase activity was performed to test this hypothesis. Gene of the luciferase is contained in the used luciferase vector. The short and the long part of the DLX1 promoter (around AP-1 site) were inserted before the gene of the luciferase in the constructs used in this method. The results of this study indicate that the regulation of gene expression through AP-1 promoter binding site is important but not sufficient part of the regulatory cascade running through ERK and JNK pathway.
