[cze][eng] Látky působící jako nekompetitivní antagonisté NMDA receptorů mohou mít psychomimetický efekt - jsou schopné vyvolat u zdravých dobrovolníků chování podobné schizofrenii. Jednou z látek s těmito vlastnostmi je i dizocilpin (MK-801), který se často využívá k modelování chování podobného schizofrenii u experimentálních zvířat ve farmakologickém modelu schizofrenie. Krom toho mají ale nekompetitivní antagonisté NMDA receptorů prokazatelné antidepresivní účinky u pacientů trpících depresí. Kognitivní deficit je v současnosti považován za jeden ze stěžejních symptomů schizofrenního onemocnění. Klíčovou roli by v kognitivním deficitu mohl hrát proces kognitivní koordinace, který se podílí na rozlišování mezi relevantními a irrelevantními stimuly. Užitečným nástrojem pro studium tohoto procesu by mohla být úloha aktivního vyhýbání se místu (AAPA), ve které experimentální zvíře musí rozlišovat mezi podstatnými a nepodstatnými stimuly ze dvou prostorových referenčních rámců. Cílem této práce bylo studium 1) behaviorálních strategií laboratorních potkanů v úloze AAPA a 2) vlivu MK-801 na behaviorální strategie a kognitivní výkonnost potkanů v této úloze. Během tohoto experimentu zaujali potkani v úloze AAPA dvě zcela odlišné behaviorální strategie. Potkani buď úlohu řešili aktivním vyhýbáním se averzivnímu sektoru,...Non-competitive antagonists of NMDA receptors can induce psychomimetic effects - they can cause schizophrenia-like behavior in healthy volunteers. MK-801 is such an agent. It is often used to model schizophrenia-like behavior in experimental animals. On the other hand, non-competitive antagonists of NMDA receptors show antidepressant effects both in patients suffering from depression and in animal models. Currently, cognitive deficit is considered to be a crucial symptom of the schizophrenia. Cognitive coordination is a process distinguishing irrelevant and relevant stimuli. A disruption of this process could play a pivotal role in cognitive deficit in schizophrenia. Active Allothetic Place Avoidance task (AAPA) could be a useful tool to study this phenomenon. In this task an animal has to distinguish between two spatial (reference) frames, whereas one of them is irrelevant and the other is relevant. The aims of my diploma thesis were: to study 1) behavioral strategies of laboratory rats in AAPA task and 2) effect of MK-801 on behavioral strategies and cognitive efficiency of rats in this task. The rats demonstrated two different strategies in the AAPA task.
