Monocyty a makrofágy patří mezi významné složky vrozené imunitní odpovědi. Tyto mononukleární fagocyty tvoří heterogenní populace buněk, které v přítomnosti různých signálu z mikroprostředí mohou měnit svůj fenotyp a funkci. Cílem této práce byla modulace jejich fenotypu spektrem stimulans a porovnání těchto změn na THP-1 monocyto/makrofágové linii a na monocytech izolovaných z lidské periferní krve. Exprese povrchových znaků byla měřena pomocí průtokové cytometrie. Dále byla analyzována produkce IL-8 metodou Luminex a koncentrace solubilního kalprotektinu pomocí metody ELISA. K nejvýznamnější změně exprese povrchových znaků docházelo po stimulaci IFNγ. Tento cytokin indukoval povrchovou expresi molekuly CD54, CD14 a HLA-DR na THP-1 buněčné linii. Při použití IFNγ o vyšších koncentracích pak byly THP-1 buňky ve zvýšené míře apoptické a zvyšovaly expresi a produkci kalprotektinu. Na membránách monocytů IFNγ aktivoval pouze expresi CD54. Cytokin IL-4 zvyšoval expresi znaku CD36 na THP-1 buňkách a inhiboval expresi molekuly CD163 na monocytech. Působením LPS pak docházelo k potlačení aktivace molekuly HLA-DR na monocytech. Inkubací s THP-1 buňkami naopak LPS indukoval produkci IL-8. Po stimulaci pomocí TNFα docházelo k aktivaci CD54 a zvýšené produkci IL-8 THP-1 buňkami. Práce přispěla ke... Monocytes and macrophages are important components of the innate immune response. These mononuclear phagocytes form a heterogeneous cell population, of which phenotype and functions can be modified under the influence of different signals coming from the surrounding microenvironment. The aim of this work was to modulate the phenotype of these cells by a variety of stimulants and to compare the changes induced on the model of THP-1 monocytic cell line and on the human peripheral blood monocytes. Surface marker expression was analyzed by flow cytometry. Further on, IL-8 production was evaluated by Luminex assay and the concentration of soluble calprotectin was assessed by ELISA. The most significant changes in surface marker expression were induced by exposure to IFNγ. This cytokine increased the expression of CD54, CD14 and HLA-DR on the surface of THP-1 cell line. Higher concentrations of IFNγ promoted higher apoptotic rate and augmented calprotectin expression and production in THP-1 cell line. On the surface of monocytes, IFNγ stimulation resulted only in the upregulation of CD54 expression. IL-4 increased the expression of CD36 by THP-1 cell line and inhibited the expression of CD163 by human monocytes.
