(CZECH) Úvod: Výzkum použití imunosuprese v kardiovaskulární chirurgii a intervenční kardiologii je v současnosti zaměřen na 3 velké oblasti: 1) ovlivnění intimální hyperplazie koronárních i periferních žilních bypasů, 2) ovlivnění rejekce cévních allograftů, 3) ovlivnění intimální hyperplazie koronárních tepen po jejich perkutánní plastice. Jedním z moderních imunosupresivních přípravků vhodným k použití v těchto indikacích je takrolimus - FK506. Při použití v experimentu inhiboval FK506 reakci tepenných alloštěpů i rozvoj intimální hyperplazie po perkutánní plastice koronárních tepen. Cíl práce: Ověřit účinky imunosuprese pomocí FK506 na průběh arterializace syngenních a allogenních žilních štěpů u potkanů. Materiál a metody: Samce potkanů kmene Lewis (LEW) (n = 46) jsme použili jako příjemce žilních štěpů. Dárcem allogenních štěpů byli potkani kmene Brown-Norway (BN) (n = 13) a syngenních štěpů potkani kmen Lewis (LEW) (n = 10). Příjemce jsme rozdělili do šesti skupin. Ve skupině A, E a F byla zvířata po syngenní (LEW do LEW) a ve skupině B, C a D po allogenní (BN do LEW) transplantaci. Zvířata ve skupině C a F dostávala denně 0.02 mg a zvířata ve skupině D a F 0.01 mg FK506 na 100 g váhy. Skupina A a B byla bez imunosuprese. (ENGLISH) Background: The investigation of the immunosuppression usage in cardiovascular surgery and interventional cardiology is, at present, concentrated on three main topics: 1) influence on intimal hyperplasia of coronary and peripheral vascular reconstructions 2) influence on rejection of allogeneic vascular grafts and 3) influence on intimal hyperplasia of coronary arteries after endovascular interventions. Modern immunosuppressive drug FK506 (Tacrolimus) could have a positive effect for these indications. In experimental study, FK506 inhibited rejection of arterial allografts and also inhibited intimal hyperplasia in percutaneous coronary interventions. Aims: The purpose of this study was to evaluate the effect of systemic tacrolimus treatment on the process of arterialisation of allogeneic and syngeneic venous grafts in a rat vein-to-artery implantation model. Material and Methods: Lewis (LEW) rats were used as recipients of syngeneic (Lewis) or allogeneic (Brown-Norway; BN) iliolumbar veins which were implanted into abdominal aorta.
