The thesis is mapping possibilities of separation of toxicologically relevant arsenic species complexed with peptides by reversed-phase high-performance liquid chromatography-inductively coupled plasma mass spectrometry (RP-HPLC-ICP-MS). The C18 chromatography column and gradient elution of the mobile phase (0,1% formic acid with gradient of acetonitrile) was used. At these conditions the separation of simple arsenic species - arsenite (As(III)), arsenate (As(V)), monomethylarsenic acid (MA(III)), monomethylarsenous acid (MA(V)), dimethylarsenic acid (DMA(III)) and dimethylarsenous acid (DMA(V)) was not successful. However, separation of arsenic- -glutathione complexes - arsenic triglutathione (ATG), methylarsenic diglutathione (MADG) and dimethylarsenic glutathione (DMAG) was obtained. Several chromatographic peaks were observed at higher concentration of acetonitrile in mobile phase. The analysis of two in vitro methylation mixtures was also examined. Mixtures contained arsenite (As(III)), S-(5'-adenosyl)methionine chloride (SAM), tris(2-carboxyethyl)phosphine (TCEP) and arsenic(3+)methyltrasferase enzyme (AS3MT) probably participating in human arsenic metabolism. Cieľom tejto práce bolo previesť prvotné experimenty separácie toxikologicky významných špécií arzénu v komplexoch s peptidmi pomocou kombinovanej techniky využívajúcej vysokoúčinnú kvapalinovú chromatografiu na obrátených fázach v spojení s hmotnostnou spektrometriou s indukčne viazanou plazmou (RP-HPLC-ICP-MS). Pre separáciu bola použitá C18 chromatografická kolóna a gradientová elúcia mobilnej fázy, ktorá obsahovala 0,1% kyselinu mravčiu a premenné zastúpenie acetonitrilu. Separácia jednoduchých arzénových špécií - arzenitanu (As(III)), arzeničnanu (As(V)), metylarzenitanu (MA(III)), metylarzeničnanu (MA(V)), dimetylarzenitanu (DMA(III)) a dimetylarzeničnanu (DMA(V)) bola za týchto podmienok neúspešná. Lepšie výsledky boli získané separáciou zložitejších arzénových špécií - arzén triglutatiónu (ATG), metylarzén diglutatiónu (MADG) a dimetylarzén glutatiónu (DMAG), kedy sa za použitia vyššieho obsahu acetonitrilu v mobilnej fáze podarilo zaznamenať niekoľko chromatografických píkov zložitejších arzénových špécií. Bola taktiež prevedená analýza dvoch in vitro metylačných zmesí, obsahujúcich arzenitan (As(III)), S-(5'-adenozyl)metionín chlorid (SAM), tris(2-karboxyetyl)fosfán (TCEP) a enzým arzén(3+)metyltransferáza (AS3MT) pravdepodobne sa zúčastňujúci metabolizmu anorganického arzénu v ľudskom...
