Vladimír Wsól, Ph.D. Školitel specialista: RNDr. Zuzana Kročová, Ph.D Název disertační práce: Studium buněčných a molekulárních mechanismů interakce B-buněk s intracelulárním patogenem Francisella tularensis F. tularensis je Gram-negativní, fakultativně intracelulární bakterie způsobující zoonózu tularémii. Imunita vůči F. tularensis je hlavně zprostředkována T-buňkami, ale důležitou úlohu v časné fázi imunitní reakce mají také B-buňky. Hlavními hostitelskými buňkami pro F. tularensis jsou makrofágy. V makrofázích se bakterie usidluje ve fagozómu, ze kterého následně uniká do cytosolu. Zde se F. tularensis množí a indukuje apoptózu a později nekrózu hostitelských buněk. Vliv B-buněk na časnou imunitní reakci proti F. tularensis je studován teprve v posledních letech. Elkins a spol. prokázali silnou časnou protekci u myší závislou na B-buňkách. Přímá interakce B-buněk a F. tularensis zatím nebyla popsána. Během naší práce jsme pozorovali adhezi F. tularensis na lidské a myší B-buňky, detekovali jsme bakterie uvnitř buněk, většinou v membránami uzavřených vakuolách a následně jsme pozorovali morfologické změny mitochondrií buňky. Immunity to F. tularensis is largely mediated by T lymphocytes but an important role of B lymphocytes during early stage of infection was previously uncovered. The target host cells for F. tularensis replication are macrophages. After the entry into macrophages, F. tularensis resides in phagosomes and then escapes into the cytosol. It replicates within the cytosol and leads macrophages to apoptosis. Recently, important role of B lymphocytes in innate immunity against F. tularensis was demonstrated. Elkins et al. demonstrated strong early protection of mice that was highly dependent on B cells. Direct interaction of B lymphocytes with F. tularensis has not been described.
