RNDr. Lenka Skálová, Ph.D. Title of diploma thesis: Cytotoxicity of new taxol derivatives in selected tumor cell lines. Paclitaxel (Taxol, PTX) is a natural product with antitumor activity, used in the treatment of various types of solid tumors, especially breast and ovarian cancer. It belongs to the so-called mitotic poisons, which cause cell cycle arrest and apoptosis of cells by interference with microtubule dynamics. The aim of this study was to compare the antiproliferative effect of PTX and its derivatives, prepared in the ÚEB AVČR by conjugation of peptide receptor agonists for gonadotropin-releasing hormone (GnRH) within a targeted cancer therapy. For the experiment was chosen cell line MCF-7, which is derived from breast adenocarcinoma and has receptors for GnRH. The results were evaluated by two different methods (NRU, MTT). Of the six tested substances showed derivative MP 265 the most significant antiproliferative effect. Paclitaxel (Taxol, PTX) je prírodný produkt s antitumorovu aktivitou, využívaný v terapii rôznych druhov solídnych nádorov, predovšetkým prsníkov a vaječníkov. Patrí k tzv. mitotickým jedom, ktoré spôsobujú zastavenie bunkového cyklu a apoptózu bunky, zásahom do dynamiky mikrotubulov. Cieľom tejto práce bolo porovnanie antiproliferatívneho účinku PTX a jeho derivátov, ktoré boli pripravené v ÚEB AVČR konjugáciou s peptidovými agonistami receptoru pre hormón uvolňující gonadotropíny (GnRH), v rámci cielenej protinádorovej terapie. Pre experiment bola zvolená bunková línia MCF-7, ktorá je odvodená z prsného adenokarcinómu a má receptory pre GnRH. Výsledky boli hodnotené dvoma rozdielnymi metódami (NRU, MTT). Zo šiestich testovaných látok vykazoval najsignifikantnejší antiproliferatívny efekt derivát MP 265. Pri použití ostatných látok nastával výraznejší pokles životnosti buniek v porovnaní s PTX až pri vyšších koncentráciách.
