Předkládaná diplomová práce se zabývá studiem voltametrického chování sulfamethizolu na uhlíkových pastových elektrodách. Sulfamethizol je sulfonamidové antibiotikum účinné proti řadě gram-pozitivních i gram-negativních bakterií. V medicíně se nejčastěji používá při léčbě močových cest. Z veterinárního hlediska se používá nejen k léčbě, ale zejména jako doplněk stravy jak vodních, tak suchozemských živočichů. Vzhledem k širokému použití a minimální metabolizaci organismem představuje také zátěž pro životní prostředí. Proto je nutný vývoj rychlých a citlivých metod pro stanovení těchto látek v biologických matricích i v lékových formách. Tato diplomová práce se zabývá nalezením vhodných podmínek pro stanovení sulfamethizolu v tabletě léčiva Micturol sedante, určeného k léčbě močových cest, pomocí diferenční pulsní voltametrie. Metoda využívá prostředí B-R pufru pH 7 a dosahuje meze detekce 0,39 µmol·l-1 . Získané výsledky byly ve shodě s údaji výrobce i s výsledky spektrofotometrického stanovení. Následně byly optimalizovány podmínky pro stanovení sulfamethizolu v moči. Nalezené podmínky byly: Lichrospher® RP-18, 100 (5 µm), 125×4 mm, (LichroCART), mobilní fáze B-R pufr pH 3 : methanol (7:3, V/V) a detekční potenciál +1,3 V. Získaná mez detekce byla 0,041 µmol·l-1 . K úpravě vzorku před samotným... The proposed Thesis deals with the electrochemical behavior of sulfamethizole on carbon paste electrodes. Sulfamethizole is a sulfonamide antibiotic effective against a number of gram-positive and gram-negative bacteria. In human medicine, the most frequent application is for the treatment of urinary tract. In veterinary medicine, it is used for the and preventive treatment of both aquatic and terrestic animals. Due to the wide utilization and low metabolization, it also presents an environmental burden. The development of quick and sensitive methods for the determination in both biological and pharmaceutical matrices is therefore important. This Thesis deals with the search of optimum conditions for differential pulse voltammetric determination of sulfamethizole in tablets Micturol sedante, used for the treatment of urinary tract. The method uses Britton-Robinson (B-R) buffer pH 7 and reaches detection limit 0,39 µmol·l-1 . Results of the method complied with the manufacturer's information and with the results of spectrophotometric determination.
