Signální MAPK (mitogen-activated protein kinase) kaskáda představuje evolučně konzervovaný mechanismus, který buňkám umožňuje vnímat extracelulární signály a vhodným způsobem na tyto signály reagovat. Nejlépe prostudovaným členem MAPK rodiny je proteinkináza ERK (extracellular signal-regulated kinase), která spolu s proteiny Raf a MEK (MAPK/ERK kinase) tvoří prototypickou signální dráhu regulující široké spektrum biologických dějů jako je kupříkladu proliferace, diferenciace, buněčný pohyb, adheze či apoptóza. Aby mohla jedna signální dráha regulovat množství různých odpovědí, vyvinuly si buňky modulační mechanismy, mezi které patří odlišná síla a doba trvání signálu, různá lokalizace proteinů, komunikace s ostatními signálními drahami, rozdíly ve výběru substrátů i to, jakým způsobem si signalizační molekuly dané spektrum interakčních partnerů vybírají. Uvedené mechanismy jsou ve velké míře ovlivňovány proteiny s neenzymatickými funkcemi, jako jsou tzv. scaffold proteiny, proteinové inhibitory a proteinové kotvy. Tyto proteiny usměrňují signály vedoucí k dosažení konkrétní buněčné odpovědi a jsou tedy klíčovými články signální transdukce. I přes rostoucí význam proteinových modulátorů v buněčné signalizaci, není jejich fyziologická role často známá. Částečnou výjimku představují dva fyziologické děje -... The MAPK (mitogen-activated protein kinase) cascade represents an evolutionary conserved mechanism by which cells sense extracellular signals and convert them into variety of context-dependent responses. The best studied member of the MAPK protein family is protein kinase ERK (extracellular signal-regulated kinase). Together with protein kinases Raf and MEK (MAPK/ERK kinase) comprise a prototypical signaling pathway which regulates broad-spectrum of biological processes such as cellular proliferation, differentiation, cellular migration, adhesion or apoptosis. To modulate such a multitude of distinct responses by a single pathway, cells utilize mechanisms such as signal strength and duration, distinct protein localization, communication with other signaling pathways, differential substrate selection and the selection of interactive partners. All presented means of regulation are influenced by proteins with non-enzymatic functions - scaffold proteins, protein inhibitors and anchoring proteins. These protein modulators channel the signals leading to particular cellular response, and thus represent the key element of signal transduction.
