5 Abstrakt Tato diplomová práce se zabývá přípravou potenciálních nefluoreskujících zhášečů fluorescence. Tyto zhášeče jsou odvozeny od nesymetricky substituovaných alkylaminoderivátů tetrapyrazinoporfyrazinů - azaftalokyaninů. Aby mohly nové sloučeniny vzniknout, bylo třeba si předem připravit podle již známých postupů potřebné prekurzory např. 2,3-dikyanchinoxalin-6-karboxylovou kyselinu. Konečné produkty byly syntetizovány cyklotetramerizací, pomocí metody statické kondenzace, ke které byli jako výchozí látky použity dva strukturně rozdílné prekurzory. V této diplomové práci byly jako výchozí látky zvoleny N-(3-azidopropyl)-2,3- dikyanchinoxalin-6-karboxamid spolu s 5,6-bis(diethylamino)pyrazin-2,3- dikarbonitrilem a ty byly cyklizovány v poměru 1:3. Cílem bylo vytvořit finální látku, obsahující periferní funkční skupinu, kterou byl zvolen azid. Sloučeninu se bohužel touto cestou připravit nepodařilo. Po dvou neúspěšných pokusech bylo patrné, že touto cestou to nepůjde a byla tedy vyzkoušena cyklotetramerizace s původní kyselinou 2,3-dikyanchinoxalin-6-karboxylovou. Ani tato cesta ovšem nebyla úspěšná, sloučenina se po přidání azidu rozpadla. Dále byla na periferii již dříve na katedře připravené kyseliny Q18-1H2 přidána periferní skupina azidu a centrálně komplexován atom zinku. Získané látky byly... 6 Abstract This diploma work deals with preparation potentially non-fluorescent quenchers of fluorescence. These quenchers are derived from asymmetrically substituted alkylaminoderivatives of tetrapyrazinoporfyrazines - azaphthalocyanines. We had to prepare necessary precursors, for example 2,3-dicyanoqinoxaline-6- carboxylic acid, by known reaction methodologies, for synthesis of new compound. Final products were synthetised by cyclotetramerization reaction and using statistic condensation method. Two structural different precursors were used as initial substances. As two initial substances N-(3-azidopropyl)-2,3-dicyanoqinoxaline-6- carboxamide and 5,6-bis(diethylamino)pyrazine-2,3-dicarbonitrile were selected in this thesis. These two products reacted in ratio 1:3. The aim was to prepare final substance with peripheral functional moieties. Azide group was selected as peripheral moiety . This synthesis was not successful. Two unsuccessful trials for preparation this substance showed, that we have to take the other way. Therefore, the 2,3-dicyanoquinoxaline-6-carboxylic was used for the cyclotetramerisaion reaction.
