Abstract: The main objective of this work is to define and characterize point mutations of the calcium channel Orai isolated form the large scale cancer genomic dataset. Those mutats were examined in the means of sub-cellular localization as well as functionality. Out of 17 investigated mutations, 5 were found to alter protein gating and switch the channel to partially or fully open state. Altered gating then leads to impaired signalling and uncontroled transcription.
