V diplomové práci byl sledován průběh kvašení ve spilce vybraného pivovaru, závislosti různých faktorů a jejích případný vliv na vitalitu a viabilitu nasazovaných kvasnic, což mělo ukázat opodstatněnou vícenásobnou použitelnost kvasničného kmene v rámci správného technologického procesu kvašení piva. Předmětem sledování byli i další faktory: fyziologický stav kvasnic, pH, teplota, množství jemných a hrubých kalů, mnpožství rozpuštěného kyslíku v mladině, stupeň prokvašení mladiny. Ke stanovení se odebíraly vzorky kvasící mladiny a vypraných násadnéch kvasnic. Použitý kmen násadních kvasnic byl sledován od jeho prvního nasazení až po poslední (celkově byl nasezen 4x po sobě). U odebraných vzorků mladiny bylo vždy změřeno pH, teplota, zdánlivé prokvašení (průměrně od 4. dne od zakvašení) a počet buněk.Před každým dalším nasazením byly kvasnice vyprány a byla stanovena jejich vitalita. Metodou vitálného barvení se sledoval podíl mrtvých buněk v suspenzi kvasnic určených pro další nasazení. Množství mrtvých buněk bylo vždy pod mezní hodnotou 5%, v jednom případě nebyla nalezena dokonce žádná mrtvá buňka v odebraném vzorku. Důležitým faktorem, především pro pomnožení kvasnic, bylo množství rozpuštěného kyslíku v mladině. Byly provedeny zkoušky s nedostatečně provzdušněnou mladinou. Pomocí těchto zkoušek bylo dokázáno, že při nedostatečném provzdušnění mladiny O2 se prodloužila doba hlavného kvašení v průměru o 3 – 4 dny. Byly provedeny i zkoušky s vyšším obsahem kalů v mladině, při nichž bylo dokázáno, že yšší obsah kalů, nemá významný vliv na průběh hlavného kvašení a na vlastnosti kvasnic. Cílem této práce bylo na základě zpracovaných výsledků sledovaných parametrů u opětovně nasazovaného (celkově 4x) kmene pivovarských kvasnic během hlavního kvašení, potvrdit správnost jednotlivých opakovaných nasazeních v průběhu technologického procesu.
In this diploma thesis, the properities of brewery’s yeasts during the operation beer fermentation in the selected brewery and dependences of diferent agents and their influence on viability and vitality of the yeasts were studied. Other observed properities were physiological state, pH, temperature, a value of soft and coarse sludges, a value of dissolved oxygen in wort, the level of wort’s fermentation. It were collected the samples of fermenting wort and washing yeasts for determination. The used yeasts were observed from the first time until thein last used (generally 4 times). pH, temperature, apparent fermenting and total numer of yeast cells were measured in the collected samples. The used yeasts were washed before every other application and vitality was determined. The number of death cells were observed by this method. The number of death cells never fell bellow 5 %. The value of dissolved oxygen in wort is also the important agent for the growth of yeasts. The touchs with low number of oxygen showed that the time for fermentation had to be longer (by about 3 or 4 days more). The touchs with higher number of sludges showed that the sludges haven’t any influence on the yeasts and the procces of fermentation. Confirm the accuracy of the repeated used of the yeasts was the object of this thesis.
