Use of Ingressive Speech Forms in Conversational Norwegian
Vaňková, Markéta ; Zíková, Magdalena (advisor) ; Friedová, Mirjam (referee)
The subject of this bachelor thesis is ingressive pulmonic speech (IPS) in conversational Norwegian. This applies particularly to verbal forms of agreement (ja, jo, okay etc.) and disagreement (nei etc.). In contrast to previous studies, the goal of this thesis is a complex and quantifiable analysis of IPS. The data consists of 60 items of IPS from a Norwegian talk show called Lindmo. In this talk show the presenter and some of her guests use these forms rather frequently. Each item is classified according to a set of parameters, including phonetic (strength, presence of F0 and intonation) and discourse features (the (macro)theme (character of the conversational topic), the position in the (macro)thematic unit, the signalization of turn-taking, pauses before/after IPS, overlaps, discourse function and the type of an utterance before IPS). All occurrences are evaluated one by one using each of the parameters. The following findings emerged from the analysis: (i) IPS are used in the media by professional speakers (the presenter), in contrast to the expectations of previous studies; (ii) most of IPS (65 %) occurs in the middle of the thematic unit, i.e. in its centre, in contrast to the claim of previous studies that one of the main functions of IPS is the closure of the thematic unit (which is only...
Study of Common Pathogenetic Factors of Alzheimer Disease and Type 2 Diabetes Mellitus
Vacínová, Gabriela ; Vaňková, Markéta (advisor) ; Lukášová, Petra (referee)
Alzheimer's disease (AD) and type 2 diabetes mellitus (T2DM) are aging-associated diseases that have rising prevalence in all industrialized countries. AD is a neurodegenerative disease characterized by progressive loss of cognitive functions. It is a complex disease which formation involves both genetic factors and environmental factors. The most important marker associated with this disease is the risk allele ε4 in APOE gene. From the latest genome-wide association study emerged another ten candidate genes. As the most significant from those genes appears the minority G allele of rs744373 polymorphism in the gene BIN1. AD is connected with many metabolic and immune disorders. To the markers of interest belongs also the new parameter visfatin which can act as a pro-inflammatory cytokine. T2DM is a chronic disease characterized by raised levels of blood glucose, which is also characterized by neurological disorders. In the case of both of these diseases can be found a large number of metabolic disorders. One of the most important disorders is insulin resistance. This thesis consists of two parts - the biochemical and genetic one. The biochemical part of the thesis studies the visfatin level in patients with AD and healthly control and studies whether visfatin is related to AD. In this part of the...
Genetic and Hormonal Regulation of Children's Growth
Vosáhlo, Jan ; Lebl, Jan (advisor) ; Marek, Josef (referee) ; Vaňková, Markéta (referee)
Genetic and Hormonal Regulation of Children's Growth MUDr. Jan Vosáhlo Abstract Growth in childhood is a complex process of changing the body, which can be disrupted by various illnesses including endocrine disorders, particularly growth hormone deficiency. Tumors or other processes affecting hypothalamic-pituitary area can be a postnatal cause of GHD; prenatal causes include 1) developmental disorders of the pituitary as part of complex syndromes, 2) developmental disorders of the pituitary due to defects in regulatory genes and 3) defects in genes involved in the synthesis and secretion of GH. The first topic of the thesis was septo-optic dysplasia - a complex syndrome involving optic nerve hypoplasia, structural brain abnormalities and pituitary dysfunctions. We extensively described phenotype in 11 Czech patients; we observed both complete SOD and incomplete forms variously combining two of the three main components of the syndrome. The cohort then became a part of an international study of 68 patients, in which we studied the phenotype in dependence on the brain morphology. We found correlation between the severity of clinical symptoms and the degree of septum pellucidum abnormities and also a correlation between hippocampus and falx abnormities and neurological symptoms. As the second topic we studied...
Study of candidate genes for obesity in children and adolescents.
Dušátková, Lenka ; Zamrazilová, Hana (advisor) ; Vaňková, Markéta (referee)
Obesity is one of the most common metabolic diseases in the whole world that increases risk of many other health problems. Origin of this multifactorial disease is influenced by many factors, including genetic ones. This work was realized within the project COPAT (Childhood Obesity Prevalence And Treatment). The main goal was to study two single nucleotide polymorphisms (SNPs) (rs9939609, rs1421085) in FTO (fat mass and obesity associated) gene and their possible impact on anthropometric traits, body composition or weight reduction. We examined two groups - 356 overweight and obese children who took part in 4-weeks weight reduction program and 412 Czech adolescents of the control group. Genotyping was performed using TaqMan allelic discrimination. Weight reduction program resulted in significantly loss of most of the examinated antropometric traits including body fat mass and visceral fat (p < 0,001). Results showed significant association of the minor A-alelle of rs9939609 with overweight and obesity (OR = 1,33; 95%CI 1,08−1,64) as well as of the minor C-alelle of rs1421085 (OR = 1,38; 95%CI 1,12−1,70). Nevertheless, there was no clear evidence of interaction between the FTO rs9939609 genotype and antropometric traits, body composition or weight loss. Results of this work confirmed that tested...

Vaňková, Markéta ; Uherek, Zdeněk (advisor) ; Moravcová, Mirjam (referee)
The thesis results from a qualitative anthropological research conducted in the border area of Šluknovský výběžek. The research subject is the construction and representation of local identites by the inhabitants of the borderland. The first part of the thesis contains an analyzis of local identity construction by the people who migrated into the studied area in the 2nd half of the 20th century. Furthermore, there are described distinct post-migration adaptation strategies of the new settlers. In the second part the author focuses on the public representations of the studied area by local activists. Three different projects aspiring to represent and thus establish the studied area as an internally homogenous (trans-border) region are analyzed. Thereby the thesis refers to the issue of the European Union and interconnected idea of European regionalism.
