Determination of selected hormonal drugs using liquid chromatography with mass detection

This master’s thesis is focused on optimizing of the analytical method for the determination of selected drugs - estrone, estradiole, ethynylestradiol, estriole, diethylstilestrole, norethindrone, mestranole and progestrone in wastewater using liquid chromatography with mass spectrometry (LC/MS). Solid phase extraction (SPE) using Oasis HLB cartridges was chosen extraction method Different mobile phase, temperature, flow rates and mobil phase gradient were during the process of optimization. Samples of the wastewater were collectedat the inflow and outlow of the wastewater treatment plant in Břeclav and in University of Veterinary and Pharmaceutical Sciences Brno. Drugs were isolated from wastewater and preconcentrated using solid phase extraction (SPE) and determined by high performance liquid chromatography with mass spectrometric detection (HPLC/MS).
Evaluation of environmental load by perfluorinated compounds

The most important substances of the group of perfluorinated compounds are perfluorooctyl sulfonate (PFOS), perfluorooctanoic acid (PFOA) and perfluorooctane sulphonamide (FOSA). Perfluorinated compounds have special physical and chemical properties, which make them valuable for usage in different industrial branches. These properties, which make them so useful are also the reason, why they are so harmful for the environment. The target of theoretical part of the diploma thesis was processing of literature searche on the topic: Evaluation of environmental load by perfluorinated compounds. In the experimental part was developed and optimized a method that was used for the identification and quantification of selected pollutants contained in real soil samples.
Assessment of the suitability of thermal water for the relaxational- therapeutic application

My thesis is based on an assessment of the suitability of thermal spring in the Southern Moravia for a relaxing, therapeutic purposes. By capillary electrophoresis are measured concentration of natural substancies, which are used in thermal waters and are searched for therapeutic effects.
Determination of selected musk compounds in biotic samples

Musk compounds (MUSK) or synthetic fragrances are organic substances commonly used as fragrant constituents of detergents, soaps, cosmetics, personal care products, industrial and in-house cleaning agents, industrial plasticizers, chewing tobacco and fresheners. The big attention is pushed ahead studying these compounds, their fate in different parts of ecosystems and studying their characteristics at present, because musks infiltrate many environmental components, particularly aquatic and marine ecosystems, through their large application and their ability to be perzistant. This diploma thesis was focused on four synthetic “classical” fragrances used over the world. The aim of this study was to perform a method optimisation for the determination of selected fragrances in biotic matrix. There was made an evalution of ability of selected water treatment plant to clear away musks from water, these results were used for evaluating contamination measurement of aquatic ecosystem. The identification and quantification of analytes was carried out by high resolution gas chromatography - mass spectrometry (HRGC/MS).
Study of degradation of biocompatible copolymers

This diploma thesis is focused on biocompatible polymers degradation study. Copolymers were studied based on poly(lactic-co-glycolic) acid and poly(ethylene glycol) PLGA-PEG-PLGA and further these copolymers modified with itaconic acid ITA-PLGA-PEG-PLGA-ITA. This paper investigated the influence of pH phosphate solution on the degradation of polymers. Degradation of polymers occurred at 37 °C in phosphate solution with pH 4.2, 7.4 and 9.2. High performance liquid chromatography with UV-VIS detection of diode-array type was used for quantitative determination of lactic acid and glycolic acid as the final degradation products. For qualitative identification of additional degradation products were used tandem connection liquid chromatography and mass spectrometry. Gel permeation chromatography with refractive index detector was used to determine the molecular weight decrease polymer chain after the degradation.
Questions of pollutants determination in waste waters

In this bachelor thesis of theoretical character is being solved the problem of the presence of harmful substances in waste waters. Concerning inorganic substances attention is focused to lead and cadmium and their compounds. From the wide row of harmful organic compounds are chosen surfactants, polycyclic aromated hydrocarbons and polychlorinated biphenyls. There are described their chemical properties, origin and appearance in waste waters. This work also includes their analytical determination by mobile analysis and confirmatory methods. The last chapter includes standard operating procedure of the confirmatory fotometric method of determination of anionic surfactants, which is suitable for accredited laboratories.
Assessmant of surfactants levels in waters

The present Bachelor Thesis deals with problem of water contamination by synthetic surfactants. Surfactants represent the main component of washing and cleaning agents, from which they enter waste waters. The goal of Thesis was to monitor and evaluate volume of surfactants in waste waters from waste water treatment plant VFU Brno. Samples were being taken in the course of 7 days and we have monitored volume of anionic and cationic tensides in samples from influent and effluent stream of waste water treatment plant. For qualitative and quantitative determination were used mobile analytic measurement devices with photometric detection by company Merck. Results were processed and interpreted in data evaluation section in the end of this Bachelor Thesis.
The presence of pollutants in matrices of bioindicators

This master‘s thesis concerns with occurrence of heavy metals in the environment, mainly in the matrices of bioindicators. Special attention is given to occurrence of mercury in the environment and its quantification in bioindicators. Mercury and its compounds are today considered as one of the most significant contaminants in the environment. Some sorts of mushrooms cumulate sizable amounts of mercury in their fruiting body and therefore we can use them to consider loading of different areas. Eight areas in Vizovice and its surroundings were chosen for these purposes and during the years 2006 – 2008 harvested 50 samples from 19 sorts of mushrooms from there. The concentrations of Hg were determined in all samples by single-purpose absorption spectrometer AMA 254. Comparison of the mercury content between individual sorts of mushrooms in the same area as well as comparison of loading of areas was performed and also bioaccumulation ability of individual sorts of mushrooms was determined.
Water utilization in ZOO Brno

Study analyses contemporary condition of water distribution in ZOO Brno with respect to environmental defensibility of local flora and fauna. Networks and condition of single water mains has been charted. Possible ways of supplying ZOO with process water has been considered. In the second part study analyses water system, quality of water and its influence on animals kept in exposition Tropical kingdom and in pavilion of arctic wolf and american beaver.
Determination of large polyaromatics in waste waters of DEZA factory

Large polycyclic aromatic hydrocarbons with molecular mass exceeding 278 Da are rarely object of study, in spite of the fact that some compounds from this group are proved carcinogens. These compounds belong to the group of persistent organic compounds and they are considered as dangerous environmental contaminants. Diploma thesis is focused on the evaluation of the occurence of large PAHs in technological and waste waters produced in black pitch processing in the DEZA Valašské Meziříčí factory. The method of their analysis based on reversed-phase liquid chromatography with non-aqueous gradient; UV-VIS and fluorescence detection will be used. This method will enable the study of changes of large PAHs distribution at various stages of waste water treatment process.
