Diagnostics of septic condition - blood culture. Comparison of costs of clasic blood cultures and automated blood culture systems
Plšková, Martina ; Vejsová, Marcela (advisor) ; Kopecký, Otakar (referee)
Author: Martina Plšková Title: Diagnostics of septic condition - blood culture. Comparison of costs of clasic blood cultures and automated blood culture systems. Bachelor's work Charles University in Prague, Faculty of Pharmacy in Hradec Králové Field of study: medical laboratory technician The aim of the study: The aim of the study was to outline the issue of septic affections, to find out a percentage representation of bacterial species in positive blood cultures and to evaluate the costs of blood cultures processing. Methods: Economical comparison of two methods of blood cultures processing - the traditional manual method and the automated blood culture system Bactec 9050. Results: The result of the examination was that samples processing using the automated blood culture system Bactec 9050 is benefitial not only from the economical point of view and that the representation of bacterial species in positive blood cultures is consistent with the results of other authors. Conclusions: Automated blood culture systems bring improvement of diagnostics of septic affections and accelerated identification of bacterial agents. This is important for early initiation of antibiotic therapy.
Effect of chemotherapy on the oral microflora and the intestinal microflora in cancer patients.
Plšková, Martina ; Vejsová, Marcela (advisor) ; Kopecký, Otakar (referee)
Author: Bc. Martina Plšková Title: The effect of chemotherapy on the microflora in the oral cavity and intestinal microflora in cancer patients Field of study: Specialist in laboratory methods Objective: The aim of this study is to assess and compare changes of side effects of chemotherapy on the microflora in the oral cavity and large intestine in patients with colorectal cancer and breast cancer and to evaluate bacterial colonization during cancer treatment. Methods: Microbiological diagnostic testing of culture of swabs from the mouth and rectum. Results: The investigators found that chemotherapy has a important influence on the composition of microflora in the oral cavity and rectum. The biggest changes in the incidence of micro-organisms were observed in the Candida albicans, which is parcipitated for mucositis. Conclusions: Chemotherapy can damage the mucous membrane as in the oral cavity and in the intestine, resulting in an imbalance in the bacterial colonization of mucous membranes. This leads to an increased representation of conditionally pathogenic bacteria that are normally suppressed by the natural microflora.
The systemic inflammatory response of the organism to Clostridium difficile infection
Plšková, Martina ; Szotáková, Barbora (advisor) ; Kopecký, Otakar (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Martina Plšková Supervisor: prof. Ing. Barbora Szotáková, Ph.D. Title of rigorous thesis: The systemic inflammatory response of the organism to Clostridium difficile infection Clostridium difficile is currently considered to be the major etiological agent of healthcare-associated diarrhoea in the developed countries. CDI (Clostridium difficile infection) is a potentially life threatening disease of increasing incidence. Despite worldwide research of new highly potent drugs, CDI treatment remains a major problem with regard to the high probability of recurrence and treatment resistance. The aim of this thesis is to find out the number of patients with CDI in a given time period and how CDI affects the C-reactive protein (CRP), leukocyte and neutrophil levels. Rapid membrane enzyme immunoassay for the simultaneous detection of C. difficile glutamate dehydrogenase (GHD) antigen and toxins A and B in a single reaction was used. The RT-PCR method was applied to confirm where a positive result for antigen but negative for toxin appeared. The biochemical determination of CRP was performed turbidimetrically. Complete blood count including a differential count was performed using electrical impedance...
