The role of protein phosphorylation during progamic phase of tobacco male gametophyte development

v angličtině (English abstract) Tobacco male gametophyte has a strongly dehydrated cytoplasm and represents a metabolically inactive stage. Upon cytoplasm rehydration, pollen grain becomes metabolically active and after the activation is finished, the pollen tube growth through a selected pollen aperture starts. The rehydration together with metabolic activation are accompanied by the regulation of translation and post-translational modifications (mainly phosphorylation) of the existing proteins. In this Ph.D. thesis, there were identified phosphopeptides from tobacco (Nicotiana tabacum) mature pollen, pollen activated in vitro 5 min and pollen activated in vitro 30 min. The total proteins from the above male gametophyte stages were extracted. The protein extract was trypsinized and the acquired peptide mixture was enriched by MOAC (metal oxide/hydroxide affinity chromatography) with titanium dioxide matrix. The enriched fraction was subjected to liquid chromatography coupled with tandem mass spectrometry (LC- MS/MS). Totally, there were identified 471 phosphopeptides, carrying 432 exactly localized phosphorylation sites. The acquired peptide identifications were mapped to 301 phosphoproteins that were placed into 13 functional categories, dominant of which were transcription, protein synthesis,...

Title: Relation between throwing velocity and accuracy in handball on elementary level Goal: Throwing effectiveness in handball can be influenced by many factors. Throwing accuracy and velocity are considered to be the determinative factors. The aim of this bachelor thesis was to explore the mutual relation of these parameters in case of handball players on elementary level. The field research was realized in the handball club Lovci Lovosice in age category of 12-13 years old players. Methodology: In terms of this bachelor thesis, two field researches in the interval of two months were realised. These field researches were realized in the handball club Lovci Lovosice in the age category of 12-13 years old players by using a hand-operated radar device The Stalker Pro 2. The research was performed only for one type of throw, specifically the one-handed overarm on the spot throw. Both throwing velocity and accuracy were traced within the research mentioned above. Statistical data processing was performed in Microsoft Excel 2010 using one-tailed statistical test. Transcription factors driving periodic gene expression during the fission yeast cell cycle

The fission yeast Schizosaccharomyces pombe plays an important role in elucidation of the mechanisms of cell cycle regulation and characterization of the relevant effector molecules involved. The cell cycle of S. pombe consists of a prolonged period of growth (G2 phase), which is followed by a nuclear division (M phase), a very short G1 phase and DNA replication (S phase). Already during S phase formation of division septum occurs. Cell cycle progression is regulated at multiple levels. Although the yeast S. pombe is an extensively studied model organism, knowledge of the transcriptional network regulating progression through the cell cycle is still incomplete. Transcription factors are very important regulators of gene expression and therefore their characterization is the subject of research. At the transcriptional level, several key transcription factors have been identified that regulate periodically oscillating and interdependent waves of gene expression during the cell cycle. This study summarizes the current state of knowledge in the field of the transcriptional regulation of periodic gene expression in the fission yeast cell cycle.
