It is an essential part of several enzymes and is involved in metabolic processes in the body. Excess or lack of serum copper can lead to pathological conditions. Copper chelating agents are used to treat copper toxic effects. Flavonoids are polyphenolic substances belonging to secondary metabolites of various plants. They are part of the human diet and have a positive impact on our health. They exhibit antioxidant and anti-inflammatory effects and are able to chelate transient metals, especially iron and copper. Chelation therapy is currently used in Wilson's disease in which copper is overloaded. In the future, the chelating effects of flavonoids could be used to treat neurodegenerative diseases, cardiovascular diseases or cancer. Barbora Szotáková, Ph.D Title of diploma thesis: Environmental risks of anthelmintics Anthelmintics are drugs used in human and veterinary medicine for the treatment and prophylaxis of infectious diseases caused by parasites- helminths. They are admittedly beneficial to the treated organisms. However, their overall effect on the ecosystem has not yet been sufficiently explored. Anthelmintics, together with their metabolites, enter the environment in multiple ways. One possibility is directly through urine and excrements of treated livestock or indirectly by leakage of contaminated manure into surface and groundwater. Such pollution has an effect on various animal organisms and plants that have the ability to uptake xenobiotics and remove them through metabolic pathways. The aim of this work was to investigate the effect of antihelmintics ivermectin and fenbendazole on soybean plants (Glycine max). The effect of methylviologen on secondary metabolites production in in vitro culture of Fagopyrum esculentum, variety Bambi

10 ABSTRACT The subject of this study was to evaluate the efect of abiotc elicitor on rutn producton in callus and suspension cultures of buckwheat. The cultvar of buckwheat used for this research was Fagopyrum esculentum Moench var. Bambi, cultvated in Murashige and Skoog nutrient medium with the additon of growth regulator 2,4-dichlorfenoxyacetc acid (2,4-D) in concentraton of 1 mg/l. The elicitor used in this study was a soluton of methylviologen, 1 ml of it was added to the cultures in three diferent concentratons: c1 = 100.0 mg/100 ml, c2 = 10.0 mg/100 ml and c3 = 1.0 mg/100 ml. The elicitor was afectng the cultures for 6, 12, 24, 48, 72 or 168 hours. Afer the defned period of tme, cultures were collected, dried out and stored for further analysis of rutn content. To control samples (without elicitor treatment) 1 ml of ethanol 96% was added and they were collected afer 6, 24, 72 or 168 hours. Releasing of rutn into the nutrient medium was also investgated. Rutn content in each sample of cultures and in each sample of nutrient medium was later determined by HPLC. Any signifcant increase in the producton of rutn was not observed in this study. The maximum amount of rutn detected was 0.1 mg/g DW, thus the lowest quantty detectable, and was found in suspension cultures in three cases, afer the additon of...

Both elements have a significant effect on the correct function of organs and make part of many enzymes. They are able to accept or donate electrons - conversion between oxidized (Fe3+, Cu2+) and reduced (Fe2+, Cu+) forms. If these metals are excess in organism, they are accumulated in the cells and mediate the creation of free radicals, that destroy cell structures. This deficiency is treated with chelators, which facilitate the excretion of metals from the body. Isoflavonoids are polyfenolic substances, which can have antioxidant effects and they are involved in the scavening of free radicals. Isoflavonoids can have also a pro-oxidative effect, because they are able to reduce metal ions. In this study were tested interactions (chelation and reduction) between the metabolites of isoflavonoids (equol and desmethylangolensin) and ions of iron and copper. Eva Gorbunovová Supervisor: PharmDr. Jana Karlíčková, Ph.D. Title of Rigorous Thesis: Comparison of copper ion chelation by benzoic acids Copper is the third most widespread element in the human body. It is necessary to maintain the homeostasis of the organism, the proper functioning of enzymes, participate in various metabolic pathways. Benzoic acid and its derived acid belong to a group of phenolic compounds that are formed by the metabolic processing of flavonoids in the human body and can have antioxidant, anti-inflammatory and antitumor effects that could be used in medicine. In this rigorous thesis, the copper-chelating activity of six benzoic acids was tested at different pH values using spectrophotometric methods using hematoxylin and bathocuproinedisulfonic acid disodium salt as indicators. The top copper-chelating activity with cupric ions using hematoxylin and the bathocuproinedisulfonic acid disodium salt and copper-chelating activity with cuprous ions using bathocuproinedisulfonic acid disodium salt showed two acids, namely 2,4- dihydroxybenzoic acid and 2-hydroxy-4-methoxybenzoic acid. Poisonings with macromycetes in period 2001-2016

The aim of this diploma thesis was to process an overview of previously known knowledge about acute mushroom poisonings. Thesis draws mainly from foreign literature. Provides basic information about mushrooms, their toxins and syndromes that cause. Beside the general information, one part of the thesis is devoted to the cases and studies from around the world. They describe individual clinical cases of mushroom poisoning or deal with their epidemiological situation mostly between the years 2001 - 2016. Obtained information is processed into overview tables. Thesis is particularly focused on the situation of macromycete poisonings in the Czech Republic and contains data from specialised facilities. The most intoxications occurs after ingestion of the edible fungi species. The most common intoxications by mushrooms containing toxins are after ingestion A. phalloides, A. pantherina and Psilocybe. The most frequent reported symptoms are gastrointestinal. According to the severity, mushroom poisonings are classified as less dangerous with onset of symptoms within 6 hours and life-threatening with the onset of symptoms after more than 6 hours. Analysis of Ginger root extract. I.

Bačová, B.: Analýza zázvorového extraktu. I. [Diplomová práce] Hradec Králové 2010, Univerzita Karlova v Praze, Farmaceutická fakulta. Komerční extrakt ze Zingiber officinale 8 : 1 byl analyzován na obsah gingerolu, vody a dalších látek. Byly použity chromatografické postupy včetně TLC a HPLC. Metodou hodnocení byly postupy uvedené v US Pharmacopeia 1996 a ČL 2009. Byla potvrzena přítomnost 6-gingerolu v množství 1,17%, obsah vody 89,18% a prokázané další látky podobné gingerolům.

Isoflavonoids are a group of 3-phenylchromen-4-one phenol flavonoids, many of which are biologically active. Isoflavonoids and their derivatives are sometimes referred to as phytoestrogens, because their structure and effect are similar to steroid hormones and are fed to estrogen receptors in the body. Isoflavonoids play an important role in human food as a health promoting plant substance. Isoflavonoids and their related compounds are used in many dietary supplements and only in one registered phytopharmac (Menofem). Recently, some natural isoflavones have been identified as toxins, eg. biliatrasone, which can cause biliary atresia if infants are exposed to this substance. However, with years of research and finding of new isoflavonoids together with understanding of their mechanism of action and their structure they have gained importance. It has a significant effect on the correct functioning of important organs and it plays an important role in the transfer of electrons at the major enzymatic pathways as a prostetic group. On the other hand, excess or deficiency of copper in the human body can cause many diseases. Phenylpropionic acids are group of substances which can have antioxidant, anti-inflammatory and anticancer effects. They are involved in the scavenging of free radicals and reactive oxygen species. In this diploma thesis, I tested copper chelating activity of six phenylpropionic acids at different pHs by spectrofotometric methods using a hematoxylin and a bathocuproinedisulfonic acid disodium salt as an indicator. 3-(3,4-dihydroxyphenyl)propionic acid showed the highest chelating potential but only using a hematoxylin.
