Gene expression during oocyte-cumulus expansion and preimplantation embryo development in mammals

CONCLUSIONS Major resultsof this PhD thesiscan be summarizedas follows: The real-time RT-PCR has become a tool for detection and quantifrcation of RNA targetsandis being utilized in geneexpressionstudyin our laboratory. The gene expression proÍile of GDF9, during porcine oocytes, cumulus andgranulosacells fu vitro cultivation and in vivo cumulus oocyte expansion was assessed.The obtained partial sequence was up|oaded in EMBL/GenBanlďDDBJ databaseand gene was localized on pig chromosome2. Conhary to mouse,GDFS mRNA was detectedfor the ťrrsttime also in porcine somaticfollicular cells. During cultivation,thetranscriptgraduallydecreasedin all the samples. we studied signalling pathways involved in IGFI promotion of FSH-induced oocyte-cumulusexpansion.we found that IGFI in combinationwith FSH increased the production of HA. The expression of Has2 mRNA in cumulus cells gradually decreasedduring cultivation' our results suggest that the PIK3/AKT dépendent pathwayis involved in the promotionof FSH stimulatedsynthesisof HA in porcine OCCs. Bovine blastocystsdeveloped from fertilized oocýes under different in vitro or in vivo conditions were comparedin the expressionof threegenes(Bax, L37 and S3o/.The embryosdiffer in the abundanceof Bax transcripl the elevatedlevels were detected in blastocyst completely cultivated...

Tapered optical fibers

A paper is focused on a design and manufacturing microstructure optical fibers with a high nonlinear coefficient, which was reached by using a nonlinear glass and tapering a microstructure optical fiber section. In designed highly nonlinear fiber taper, supercontinum generation was simulated. Designed tapered microstructure optical fiber was manufactured with using constructed tapering apparatus. The tapering apparatus was also employed for producing fiber tips, as probes for pH measurement.

The paper deals with technology of manufacturing fiber tapers that are made by heating and stretching a standard optical fiber (e.g. SMF, PCS or MOF) in a heat source. For fabrication of the fiber tapers have been designed and assembled the apparatus that is propelled with motors and controlled by a PC and other electronic equipments. Manufactured tapered optical fibers can be used e.g. in nonlinear optics, for various types of fiber evanescence chemical sensors and holography. 482

Measurement of short pulses using fiber frequency-resolved optical fating

We have reviewed and implemented the method of frequency-resolved optical gating. We discuss the fiber FROG experimental setup and several algorithms for pulse retrieval. These include the basic algorithm, the generalized projections and the genetic algorithm. We have developed new variants of the pulse retrieval algorithms by combining the generalized projections and the genetic algorithm and are experimenting with them to get optimal performance.

Microstructure optical fibres

A review of microstructure optical fibers, their use and methods of preparation is presented. Two classes of microstructure optical fibers in accordance with wave guiding mechanism are introduced. Unique dispersion properties and their structure dependence are discussed. Highly nonliner fibers, large mode area fibers and highly birefringent fibers are reported. Finally, the research carried out IREE Prague is summarized.
