Optimization of human carbonyl reductase overexpression in Escherichia coli

Title: The optimization of expression of the selected human carbonyl reductases in Escherichia coli The present thesis called The optimization of expression of the selected human carbonyl reductases in Escherichia coli is divided into two parts. The first one is theoretical and the second one is experimental. The theoretical part presents a brief information summary about selected enzymes: carbonyl reductases 1 (CBR1), quinone oxidoreductase 1 (NQO1) and DHRS2 (Hep27). It also includes a theoretical summary of laboratory methods which were used in the experimental part. The experimental part aims to optimize the express conditions of the selected enzymes in the bacterial expression system Lemo21(DE3). The plasmid pET-28b(+), which encodes the selected enzymes, is transformed by a thermal shock into competent bacterial cells Lemo21(DE3). In these cells a protein expression was carried out at different concentrations of L-rhamnose. We also purified the expressed recombinant protein NQO1 by the affinity chromatography method. Comparison of Results of Coagulation Parameters Measured by Different Methods

The present thesis deals with the laboratory determination of the functional activity of coagulation factor VIII. It is one of the coagulation factors involved in hemostasis. Its deficit or functional deficiency leads to a severe bleeding disorder - haemophilia A. The aim of the theoretical part of this thesis is to create a brief overview of the coagulation factor VIII - heredity, structure, biosynthesis, secretion and function in the organism. It also describes haemophilia A, which is a disorder caused by a lack of synthesis, functional defects or the presence of inhibitors against the coagulation factor VIII. The history of the disease, clinical classification, etiopathogenesis, diagnosis and treatment are described in this part. Laboratory methods for determination of coagulation factor VIII and its specific inhibitor are described in the last part. The practical part deals with the determination of the functional activity of the coagulation factor VIII by the commonly used coagulant method and also by the chromogenic method. How to support the convergence of socioeconomic indicators in Indonesian provinces?

The thesis looks for the answer to the question how to support the convergence in of Socioeconomic Indicators in Provinces of Indonesia. Together with literature review, the analytical part concerns the regional disparities and convergence of socioeconomic indicators in given period from 2010 to 2015. The analytical contains the analysis of betaconvergence calculated by regression line and the analysis of sigmaconvergence computed on the base of standard deviation change throughout the years. All this implies in proposal part where the suggestion to continuous convergence are specified: sustainable development, human development, capital mobility, labour force mobility.

Cílem bakalářské práce je charakterizovat postavení informačního pracovníka v dnešní společnosti. Práce vytváří souhrn základních poznatků o informační profesi a zároveň tuto oblast zpřehledňuje. První část práce vymezuje základní pojmy týkající se informační profese, popisy konkrétních informačních profesí a pojmy z oblasti informačního vzdělávání. Dále je uveden přehled oborů v rámci studia informační vědy v České republice a následné možnosti uplatnění absolventů tohoto studia. V bakalářské práci je kladen důraz na jednu ze zásadních organizací, týkajících se informační profese - Svaz knihovníků a informačních pracovníků České republiky - jeho historii, organizaci, projekty i mezinárodní spolupráci. Prostor je věnován také stručné charakteristice České informační společnosti.

Analysis of the retail network in Klatovy

The goal of this graduation thesis is to analyze the retail network related to food products in the city Klatovy and to evaluate whether the number of the retail units is appropriate for this area. Assess whether the number of retail units corresponds to the number of the population and the optimal area for Klatovy. The first part is focused on the different concepts and describes methods regional analysis. In the second part there is described the history and sights of the city Klatovy. After that follows characteristic of individual stores (retail units focused on the sale of food products), regional analysis of Klatovy and finally the evaluation questionnaire on the citizen buying habits and satisfaction with shopping in Klatovy.
