Psychospiritual crisis from the point of view of social and pastoral work

The content of this bachelor thesis is psychospiritual crisis from the perspective of pastoral and social work. In the theoretical part I describe the forms and manifestations of psychospiritual crisis and try to explain how this crisis differs from mental illness. I place the issue in the context of pastoral and social work. In the practical part there are individual interviews with pastoral and social workers that are part of the research. I have used a case study as the research design. The selection of respondents was based on a theoretical sampling of the data, which I then interpreted through content analysis. In the practical part, the research questions are asked. The results of the investigation are presented in the conclusion of the thesis. The aim of the study was to find out how the issue of psychospiritual crisis is perceived by social and pastoral workers. Social workers consider psychospiritual crisis as an important topic and feel that there is a lack of awareness of this issue in the field of social work. Most pastoral workers, especially in the Catholic Church, were seen to be uninterested in the topic related to psychospiritual crisis. Development of new molecular tools for altering gene expression in Giardia intestinalis .

Giardia intestinalis is a widespread intestinal parasite that causes diarrhea in human and other vertebrate hosts. Although, fully sequenced genome of G. intestinalis has been published, very little is known about the regulation of gene expression. Together with the tetraploid genome, this complicates the use of many common reverse genetics methods. The aim of this thesis was to develop new molecular tools that can be used to alter gene expression in G. intestinalis. For the purposes of this work, new vectors for tetracycline-inducible gene expression including T7 promoter and endogenous oct promoter were designed. Furthermore, cwp1 gene knock-out was created using CRISPR-Cas9 technology. In order to modify mechanisms of double strand break repair, expression of two key components of bacterial NHEJ pathway - LigD and Ku - was introduced into cells of G. intestinalis.

Statistical processing of the Hepatitis C virus genotype analysis results. Material and methods: In the period of 1 July 2005 - 30 June 2012, the Hepatitis C virus genotype analysis was performed in respect of 2 185 samples at the division of molecular biological diagnostics of the serology laboratory of the Institute of Clinical Biochemistry and Laboratory Diagnostics of the General Faculty Hospital and the First Faculty of Medicine of Charles University in Prague, using the reverse hybridization method. The testing results were statistically processed. Results: Of the set of 2 185 samples, 1 372 (62,8 %) originated from males and 813 (37,2 %) from females, with the average age of patients included in the set being 36,7 years. The majority of requests for the HCV genotype analysis originated from specialists in Prague (91,0 %). The genotype analysis demonstrated the highest prevalence of the 1b genotype (51,6 %), followed by the 3a genotype (22,6 %). The prevalence...
Inducible expression systems and their use in the study of parasitic organisms.

1 Abstract Inducible expression systems are systems with ability to switch expression of genes of interest on and off. Therefore, they are useful molecular tools for analysis of gene function. Nowadays, there are tens of various inducible expression systems available that differ from each other in level of regulation of gene expression, time of induction, possibilities of use, etc. This work is focused on three of them to illustrate common features of the inducible expression systems which regulate gene expression at the level of transcription. Firstly, systems based on regulation of lactose operon of Escherichia coli are mentioned. Secondly, systems which use regulatory elements of tetracycline resistance-encoding transposon Tn10 of E. coli are described. Third chapter is focused on systems regulated by agonists of ecdysone receptor. In the last chapter cases of use of inducible expression systems in the study of parasitic organisms are summarized.

Socio-economic impacts of the strategic industrial zones on the development of the region

The aim of this thesis is to define socio-economic impacts of the strategic industrial zones on the development of the region. The aim of the thesis was chosen with respect to the statement from literature, that industrial zones stimulate economic activity through making new jobs. In the first part of work are described teoretical fundamentals present by Czech and also foreign authors, who are focused on industrial zones problematics, characteristics of industrial zones in the Czech Republic and localization. In the second part are two case studies of strategic industrial zones Most-Joseph and Ostrava-Mošnov. The component of case studies is analysis of statistics for the period from 2000 to 2012, which results from observation of casual relations between development of zones and their surrounding areas lead to identification of economic and social impacts of strategic industrial zone on the development of the region.
Trafficking in human beings as a way of strengthening the market competitiveness of the Czech Republic?

The thesis considers trafficking as a way of strengthening the market competitiveness of the Czech Republic. Contrary to wide-spread approach to this issue, only as to violation of the rights of victims, trafficking is examined from the side of entrepreneurs and from the side of impact on national competitiveness. In addition to factors affecting the national economy and the motivation of entrepreneurs is evaluated the efficiency of subsidies provided by the Ministry of the Interior to Programme of the support and protection for victims of trafficking. In the thesis is found out that the exploitation of trafficked persons increases competitiveness of the Czech Republic, because it fills gaps on the domestic labour market with no wage increases. For employers victims are flexible and low-cost source of labour force, which increases productivity of their firms, but they don't represent significant competition for legal workers in the country.
