Multifunctional Centre of Civil Activities

The masters thesis dealt with reconstruction of the complex military garages built in the mid 20th century. The site is located in Brno, close to Peace Square, Lerchova 915/62, Brno-Stránice, 602 00 Brno. Area of land area is 5,384.25 square meters. Reconstruction involves two detached single-storey buildings, the street Lerchova object and the object at the Valley Street. The goal is the maximum possible use of existing quality substructure, which is a combination of internal reinforced concrete frame and brick outer shell. The work is divided into a comprehensive layout of both buildings and detailed sub-project for the construction project on a subject at street Lerchova. Reconstruction of the resolves completely change the layout and especially the concept of architectural design. In terms of architecture, wider relations into account the existing development, which tries to resemble the representation. The building is at street Lerchova basement one floor, built up area is 620.5 square meters. Reconstruction of superstructure solves one floor, making new pitched roof and building a walkway that allows connection between the two objects. Revitalization is due on the requirements of the public and addresses the widespread use of indoor and outdoor spaces as much as possible. It is particularly emphasized promote leisure activities not only public, but especially young people and support for education, mental and physical, education and counseling. We can not forget easily to most complex space by persons with limited mobility.

Restaurant with Point of View

The aim of the master thesis „ Restaurant with a view“ is project for realization of a building. The building is situated in city district Brno – Veveří. New building will be placed instead of a current garage in the end of the street Rybkova. The building has two floors. The bearing system is a combination of ceramic bricks Porotherm and reinforce conrete frame. The frame consists of columns and longitudinal girders. The ceiling is a monolithic reinforce concrete slab. The roofs above both floors are flat and walkable with waterproofing layer made of modified bitumen sheets. The walkable layer is made of concrete paving with a rectification targets. The projekt also solves preparation for possible placing of wooden tower on the roof of the second floor. Wooden tower was made just as initial design.

This dissertation deals with the design of the object designated as a technical service center in the town Ivančice. According to the requirements of the client the object contains the following sections: facilities for 25 people, electricians room with the storeroom, premises for the manager, the accountant and the field worker, necessary sanitary facilities, material storeroom, workshop and garages. The building is wheelchair accessible. The main vertical load-bearing constructions are built from brick blocks, the ceiling construction is monolithic reinforced concrete, the load-bearing construction of the roof is made up of wooden rafters. Workshop, garages and warehouses are located in the adjoining steel assembled hall which is sheathed in polyurethane panels.

The Financial Plan Proposal of Construction Company

The objective of this diploma thesis is to create a financial plan for a selected company. The first part deals with theoretical knowledge which is necessary for the formation of the financial plan. The second part focuses on the presentation of the selected company, and creating a strategical and financial analysis. Based on all the gathered information, two options of a financial plan will be created. The last part presents an overall evaluation of the plan.

Multifunctional building on Burian Square in Brno

The work is focused on the construction of the multifunctional building in the atypical-shaped corner area on Burian Square in Brno. The house has four floors above the ground and one underground floor. The architectural design was created according to the requirements of the city-part authority - enlarge the city road and maximize the usability of the whole parcel.

Methylation of the regulatory sequences of integrated and extrachromosomal form of HPV 16 in tonsillar tumors

Human papillomavirus is connected with induction of cervical carcinoma as well as for some other anogenital carcinomas and subset of carcinomas of head and neck. Presence of viral E6 and E7 oncoproteins may induce cell transformation, higher load of oncoproteins is caused by the regulatory E2 protein inactivation. Aims of recent study are mechanisms of E2 protein inactivation. One option is integration of viral DNA into the host genome, which is located into the E2 gene region. Some carcinomas, where virus with extrachromosomal form was presented, were found. It appears that epigenetic changes can play the role in the development of this type of tumors, especially DNA methylation or mutation in the regulatory region of the virus. The methylation degree analysis on samples of tonsillar carcinomas with extrachromosomal and integrated form of the virus was conduced, as well as viral load of both groups was compared and the expression of E6 and E7 gene was confirmed. The results of methylation analysis showed increased methylation of the virus with integrated DNA. Mutations in the E2 protein binding sites are not revealed. Detection of human respiratory DNA viruses in the respiratory tract samples of immunocompromissed patients.

Respiratory tract diseases are of the most common infectious diseases among both children and adult population all over the world. Viruses are the most frequent cause of respiratory diseases. In healthy immunocompetent individuals respiratory infection proceeds mostly without major complications. Immunocompromissed hosts, for example patients after transplatation, are more susceptible to infection and even common infection may be life threatening for them. Human polyomaviruses KI (KIPyV) and WU (WUPyV) and human bocavirus (HBoV) are most frequently detected in the respiratory tract of patients with acute respiratory tract infection primarily in children and in immunosuppressed patients. However, clear causative link between presence of these viruses and the respiratory disease has not been established. In this retrospective study were tested by quantitative real-time PCR 822 (745 from adults and 77 from children) respiratory samples from 380 immunocompromissed patients included 326 adults and 54 children. Viruses were also detected in the 84 peripheral blood samples.
