Optimization of the MRI chicken embryos measurements

This thesis deals with optimization of MRI measuring of phantoms containing the chicken embryo. The theoretical part of thesis is dedicated to the MRI fundamentals and description of pulse sequences. In the next part the options of keeping and monitoring the temperature are being discussed. The thesis also deals with design and realization of phantoms which are used for measuring. A browser of images captured by MRI was programmed in Matlab environment. The program allows users to look through the images with different settings of pulses sequences or kind of phantom. The final part of thesis describes the anatomical structures of chicken embryo and discussion of collected results.
Physiological and pathological factors affecting absorption of drugs

Absorption of drugs from the gastrointestinal tract after oral administration is a key pharmacokinetic process co-determining the subsequent pharmacodynamic response of the organism and therapeutic efficacy. This dissertation thesis is devoted to the study of factors that influence this parameter. Special emphasis is placed on the study of lymphatic absorption, i.e. the rate of absorption of the active substance via the intestinal lymphatic system. A number of in vivo studies have been carried out in laboratory rats. Pharmacokinetic studies have been performed by means of regular blood sampling from vascular catheters after oral administration of the drug. Lymphatic absorption was investigated in an anaesthetized mesenteric lymphatic duct cannulated rat model. Modern drugs were tested that were incorporated into innovative dosage forms by collaborating chemical and technological institutions. Abiraterone acetate, a lipophilic agent used in the therapy of prostate cancer, was well absorbed from the gastrointestinal tract after administration in the form of oil marbles. This technology also succeeded in reducing the otherwise very significant positive food-effect. Abiraterone acetate was not absorbed to any significant extent via the intestinal lymphatic system. Aripiprazole-induced adverse metabolic alterations in a polyI:C neurodevelopmental model of schizophrenia in rats

The aim of this study was to evaluate metabolic phenotype of poly I:C rat model and asses metabolic effects of chronic aripiprazole treatment with regard to complex neuroendocrine regulations of energy homeostasis. Altered lipid profile in poly I:C model was observed. Leptin and GLP-1 serum levels were significantly reduced, while ghrelin level was elevated. Our data indicate that dysregulation of adipose tissue endocrine function and gastrointestinal hormones is implicated in metabolic adverse effects of antipsychotics.
The influence of acute and chronic application of olanzapine on brain perfusion in rat

Olanzapine belongs to the first choice drug in treatment of schizophrenia. We focused on blood perfusion changes inducted by acute and chronic application of olanzapine in healthy animals and MAM altered animals. We detectet significant lower perfusion of cerebral cortex in acute and chronic olanzapine treated animals. We susspect that it could be due to altered neurotransmiter regulation caused by olanzapine.

A contract-based research with a specified mouse model of schizophrenia was carried out, examining the possibility of imaging relevant parameters of the structure and function of organs specified by the client. The options were assessed with respect to client's needs, suitable imaging methodology was identified and developed, animal experiment protocols were prepared and validated, and the proposed methodology was applied to a series of 60 animals according to valid animal use protocols. Original and statistically evaluated data were provided to the client.
