Praha 2008 Introduction Acyclic nucleoside phosphonates (ANPs) represent a key class of nucleotide analogs with a broad spectrum of antiviral and cytostatic activity.1 Among ANPs, particularly 9-[2- (phosphonomethoxy)ethyl]adenine (PMEA, adefovir, Figure 1) is active against DNA and retroviruses; its prodrug, adefovir dipivoxil, was approved for hepatitis B therapy (Hepsera). 9-(R)-[2-(Phosphonomethoxy)propyl]adenine (PMPA, tenofovir), is a promising anti-HIV drug, its prodrug Viread was approved for treatment of AIDS and chronic hepatitis B. A series of acyclic nucleoside diphosphate mimics bearing purine and pyrimidine bases was prepared by Mitsunobu reaction of protected heterocyclic bases with appropriate alcohol containing stable diphosphate analogue.

A new type of acyclic nucleoside phosphonates (ANP) is derived from 2,4-diamino-6-hydroxypyrimidine. These 6-[2-(phosphonomethoxy)ethoxy]pyrimidine derivatives significantly inhibit replication of retroviruses and herpesviruses in cell cultures. This novel subclass of pyrimidine ANPs can be considered as analogues of 2,6-diaminopurine with an open imidazole ring in the purine moiety.
