Chleboun, J. ; Kůs, Pavel ; Přikryl, Petr ; Segeth, Karel ; Šístek, Jakub ; Vejchodský, Tomáš
This book comprises papers that originated from the invited lectures, survey lectures, short communications, and posters presented at the 18th seminar Programs and Algorithms of Numerical Mathematics (PANM) held in Janov nad Nisou, Czech Republic, June 19-24, 2016. All the papers have been peer-reviewed. The seminar was organized by the Institute of Mathematics of the Czech Academy of Sciences under the auspices of EU-MATHS-IN.cz, Czech Network for Mathematics in Industry. It continued the previous seminars on mathematical software and numerical methods held (biennially, with only one exception) in Al šovice, Bratří kov, Janov nad Nisou, Ko řenov, L ázně Libverda, Dolní Maxov, and Prague in the period 1983-2014. The objective of this series of seminars is to provide a forum for presenting and discussing advanced theoretical as well as practical topics in numerical analysis, computer implementation of algorithms, new approaches to mathematical modeling, and single- or multi-processor applications of computational methods.

Optimization of urinary sample pre paration for proteomic analysis in kidney disease diagnosis
Juránková, Lada ; Přikryl, Petr (advisor) ; Pacáková, Věra (referee)
This work is focused on urinary sample preparation before their proteomic analysis by liquid chromatography with mass spectrometric detection. Urinary proteomics is in many cases inefficient due to low urine protein concentration, variations in pH and the presence of high amount of salts, pigments, and other metabolites. These components can interfere with proteomic protocol and with subsequent mass spectrometric analysis. Therefore, there is the need to develop new effective sample preparation method. The main benefit of this work is the optimization of the method for the effective preconcentration of proteins from urine samples using carboxylate-modified paramagnetic microparticles and the principles of HILIC chromatography. This innovative process was compared with a routine method using centrifugal filters (FASP) and the method based on proteins isolation by divinyl sulfone activated cellulose magnetic particles. Using carboxylate paramagnetic microparticles, a total of 856 proteins was identified from only 0,5 mL of healthy human urine by LC-MS/MS. This optimized protocol provided comparable results to the standard FASP method in a much shorter time and with a lower cost per analysis.
Optimization of urinary exosome isolation for proteomic analysis in kidney disease diagnosis
Ulrychová, Lucie ; Přikryl, Petr (advisor) ; Kučerová, Zdenka (referee)
Extracellular vesicles (exosomes) are the subject of current nephrology proteomics research as they are considered as a promising source of potential biomarkers of kidney disease. This work is focused on discovery of the most appropriate procedure for the urinary exosomes isolation. We have compared already described methods, based on different physicochemical principles of isolation: hydrostatic filtration dialysis (HFD), differential ultracentrifugation, ultrafiltration through a 100 kDa filter, or sample precipitation with Total Exosome Isolation (from urine) kit. Characterization of individual isolated exosomal fractions was performed using SDS-PAGE method (presence of contaminating proteins), western blot analysis (detection of exosomal markers TSG101, alix), nanoparticle tracking analysis (NTA, vesicle size and concentration) or transmission electron microscopy (TEM, vesicles morphology). Due to the presence of contaminating proteins in urine samples, which could distort the results of subsequent proteomic assays, the conditions for the cleavage of undesirable proteins by proteinase K prior to their own isolation were optimized. It has been found that the best yield and purity of the isolated exosomal fractions were provided by a process combining HFD with differential ultracentrifugation...
The use of restrictive means at patients
Přikryl, Petr ; Šlaisová, Ivana (advisor) ; Hosák, Ladislav (referee) ; Machalová, Iva (referee)
The bachelor's work deals with patients'view of the use of restrictive means. In the theoretical part are given a brief history overview of the mentally ill and attitudes towards them, methods of treatment, ways of patients'hospitalization, separate kinds of restrictive means and indication to their use. In the empiric part are evaluated interviews with seven female patients, hospitalized in the Psychiatric Clinic in Havlíčkův Brod, that were restricted in a protective net bed. The aim was to determine the females patients' view of single phases relating to the restriction and their confrontation with documentation and the care staff's view. The patients'attitude towards the given-topic interview was helpful, they willingly answered most of the 107 asked questions. The quality probe found out that the patients themselves did not assess the single restriction negatively, they admitted its validity and its beneficial effect in most of the cases. The most contradictory were descriptions of situations leading to the restrictions.

Prikryl, Petr ; Zatloukal, Miroslav (referee) ; Starý, Jiří (advisor)
The aim of this thesis is solving the issue of introducing the Dotmaster semi-automatic dispenser into the flexible low series surface mount technology assembly of printed circuit boards. The first part of the thesis deals with the issue of introduction to the dispension of solderpastes, adhesives and packaging materials. The second part is focused on the description of the Dotmaster semi-autamatic. The last part is devoted to actual measuring and observation with the device using PD 860002 SA adhesive made by Heraeus company and SAC solder paste by Kester company. Showing the characteristics of dependecies of the shape of the dispensed adhesive and solder paste on several parameters of dispension follows.
