In addition to pregnancy planning and other non-contraceptional benefits, it also has certain risks. Hormone replacement therapy (HRT) is used primarily for treatment of estrogen deficiency in postmenopausal women. HRT is associated with a number of risks, in particularly for women 60+ of age. Analysis of spontaneous adverse drug reactions (ADRs) reporting contribute to detection of potential risks associated with pharmacotherapy, thereby increases the safety of the drugs. Except for indication for depression, they are used in the therapy of chronic pain or enuresis nocturna in children. Nowadays, tricyclic antidepressants are drug of the first choice only in some types of neuropathic pain, and in other indications they have been replaced by newer substances with a lower occurrence of side effects. The aim of this works is to find out how consumption of tricyclic antidepressants has developed and what share in utilization of antidepressants they represent. Data for this work were obtained from the State Institute for Drug Control of the Czech Republic; namely from distributors' report about supplies of medicinal products to pharmacies and healthcare facilities, sellers of selected medicinal products, other distributors and veterinary doctors. Spontaneous reporting of adverse reactions is an important source of information to identify potential risks of medicinal products. Objective: The aim of this diploma thesis is the analytical evaluation of spontaneous reports of suspected adverse reactions after vaccination with MMR vaccine registered in the database of the State Institute for Drug Control during the period 2004 to 2017. Methods: The data were analyzed in Microsoft Excel spreadsheet software using descriptive statistics methods. Analysis of the Drug Related Problems in Facilities for Seniors I. Author: Veronika Duchoňová1 Tutor: Josef Malý1 1 Department of Social and Clinical Pharmacy, Faculty of Pharmacy in Hradec Králové, Charles University Introduction and aim: The aim of the thesis was to revise the medical documentation in the aftercare health facility, to classify the identified drug related problems (DRP) and to describe the chosen DRP by the SAZE principals. Methodology: The data collection for this thesis took place in ONLP in the time period from the 27th of August to the 30th of August 2019. The aim of the auditors was a revision of phamacotherapy of all 76 patients. A complete electronic and printed medical documentation was provided. The DRP were identified based on the information about a patient and described by the SAZE principals. The DRP were reported in the electronic database according to the modified PCNE classification V5.01. All obtained data was exported to the Microsoft Office Excel in the form of tables and processed by freqency analysis. Six case studies with the detailed description of the patient and the pharmacist's analysis by the SAZE principals were demonstrated. Unless properly treated, it leads to higher mortality and morbidity rates. Early detection of such fibrillation, which is often asymptomatic, may prevent further associated complications, especially ischemic stroke, and a sudden cardiac death. Aim: The thesis focused on the feasibility of involving pharmacists in the screening of the patients with heart rhythm disorders. Further increase awareness of the possibilities and meaning of monitoring heart rhythm disorders in pharmacies and ensure that patients can verify their own heart rate at home. Objective: The objective of this diploma thesis was to evaluate the utilization of AEDs in the Czech Republic in the period from 01. 01. 2004 to 31. 12. 2018 using data from the State Institute for Drug Control (SIDC). Methods: Retrospective analysis of drug utilization data. Data on the utilization of antiepileptic drugs were received from the SIDC database and included all AEDs that were distributed to medical facilities during the reporting period. The relative comparative unit DID was calculated - the number of defined daily doses per 1000 inhabitants per day. The data on the number of residents were obtained from the Czech Statistical Office. Results: Consumption of antiepileptics has increasing trend. During the period it increased from 7.43 DID to 15.87 DID. Especially in the group of new antiepileptics an increase was observed, where 1.15 DID raised to 10.59 DID, while in the classical antiepileptic group this value decreased from 6.28 DID to 5.28 DID. Consultant: PharmDr. Eva Zimčíková, Ph.D Introduction: Selective serotonin reuptake inhibitors increase the available amount of neurotransmitter serotonin in synapses, resulting in their antidepressant and anxiolytic effect. Objective: The objective of this diploma thesis was to evaluate the consumption of SSRI antidepressants in the Czech Republic in the period from 01. 01. 2008 - 31. 12. 2018, based on the data on consumption from the State Institute of Drug Control (SIDC). Methods: The research consisted of a retrospective analysis of the SIDC database. The study included all approved SSRI in the Czech Republic. Drug Utilization Review was used in the thesis as a methodology. Drug consumption is calculated as the number of defined daily doses per thousand inhabitants per day (DID). The data on the number of residents was acquired from the Czech Statistical Office. Descriptive statistics and ATC/DDD methodology were applied. Results: The consumption of sertraline and escitalopram increased significantly during the period under review. Dual antidepressants are prescribed in treatment of depressive disorder, anxiety disorder, in pain therapy and insomnia. Besides that, there are trials to prove the efficacy of dual antidepressants in different indications. Generally, antidepressant consumption in the Czech Republic is increasing. The question is, what the real proportion of dual antidepressant consumption is. Objective: As the objective, dual antidepressant consumption was analysed. Methods: The consumption of antidepressants venlafaxine, duloxetine, mirtazapine and mianserin was performed in the period from 1 January 2008 to 31 December 2018. Synthesis and Biological Activity of Novel Natural Product Derivatives

Within the framework of this Thesis, several series of 3,4-diphenylfuranones related to both combretastatin A-4 and antifungal 5-(acyloxymethyl)-3-(halophenyl)-2,5-dihydrofuran-2-ones was prepared. Cytotoxic effects on a panel of cancer and normal cell lines as well as antiinfective activity were evaluated, and the data were complemented with tests for the activation of caspase 3 and 7. High cytotoxicity was observed in some of the halogenated analogues, eg. 3-(3,4-dichlorophenyl)-4-(4-methylphenyl)-2,5-dihydrofuran-2-one with IC50 0.12-0.23 µM, but the compounds were also highly toxic against non-malignant control cells. Interestingly, notable antibacterial activity indicating G(+) selectivity has been found in the 3,4-diarylfuranone class of compounds for the first time. Hydroxymethylation of furanone C5 knocked out cytotoxic effects (up to 40 µM) while maintaining significant activity against Staphylococcus strains in some derivatives. MIC95 of the most promising compound, 3-(4- bromophenyl)-5,5-bis(hydroxymethyl)-4-(4-methylphenyl)-2,5-dihydrofuran-2-one against S. aureus strain ATCC 6538 was 0.98 µM a 3.9 µM after 24 h and 48 h, respectively.
