ABSTAKT Univerzita Karlova Farmaceutická fakulta v Hradci Králové Katedra biologických a lékařských věd Studentka: Dominika Doubová Školitel: RNDr. Klára Konečná, Ph.D. Název diplomové práce: Využití bezobratlého zvířecího modelu Galleria mellonella pro tvorbu kvasinkových biofilmových společenství Cíl práce: Cílem této diplomové práce bylo blíže představit bezobratlý organismus, Galleria mellonella (G. mellonella), který lze v rámci vědeckého výzkumu využívat jako alternativní modelový organismus k obratlovcům. Model se jeví ekonomicky výhodnějším a morálně akceptovatelnějším, což je podstatný fakt v současné době, kdy stále roste tlak na upouštění testování zejména na obratlovcích. Experimentální část této práce se pak věnuje optimalizaci využití larvy G. mellonella jakožto modelového organismu v rámci výzkumu popáleninových zranění s fungálním infektem, vyvolaným kvasinkou Candida albicans. V rámci experimentů byly larvy využívány pro přípravu tkáňových explantů, na kterých probíhalo šetření formování monodruhového kvasinkového biofilmu. Metody: Před získáváním tkáňových explantů byly larvy usmrceny dekapitací. Za účelem omezení kontaminace tkáňových vzorků především střevní mikrobiotou larev byl optimalizován přístup odstranění trávicí trubice. Popáleninové rány byly indukovány post mortem pomocí... This model seems to be more financially favorable and more ethically acceptable, which is an essential fact in the current era of increasing pressure to abandon testing on vertebrates in particular. The experimental part of this thesis is focused on optimizing the utilization of the G. mellonella larvae as a model organism within the research of fungal burn wounds fungal infections induced by the yeast Candida albicans. Within these experiments, larvae were used to preparation of tissue explants on which the formation of a single- species yeast biofilm was investigated. Methods: Before obtaining of tissue explants, the larvae were decapitated.
