Tato bakalářská práce v rešeršní části uvádí do problematiky bioaerosolu, popisuje význam pylu, způsoby jeho šíření a vliv na zdraví člověka, shrnuje metody odběru pylu z ovzduší a běžné způsoby identifikace pylových zrn. V experimentální části práce prokazuje vhodnost využití prvního stupně vysokoobjemového kaskádního impaktoru (Hi-Vol) BGI-900 k odběru biaerosolu a následné SEM analýze pylu. Byla vypracována metoda separace pylových zrn z polyuretanové pěny (PUF), která je impakčním substrátem HiVol impaktoru, na homogenní depozit na nylonový filtr. Reprezentativní část depozitu byla analyzována skenovací elektronovou mikroskopií - SEM. Pro celkem 12 vzorků ze třech lokalit v ČR, bylo vytvořeno celkem 485 snímků SEM. Na 295 snímcích byla identifikována pylová zrna do 9 rodů a 4 čeledí a kvantifikována četnost deformit, která činila 24, 18, 50% na lokalitách Praha, Březno a Láz. Pouze v Praze počet pylových zrn rostl s celkovou hmotností aerosolu. Ve vzorcích byly identifikovány sekreční produkty hmyzu brochosomy. This bachelor thesis, in literature review, deals with bioaerosol issue, describes meaning of pollen, ways of pollen spreading and its influence on human health, summarize methods of pollen sampling from the air and common ways of pollen grains identification. In the experimental part, the use of the first stage of a High Volumetric Cascade Impactor (Hi-Vol) BGI-900 for bioaerosol sampling and subsequent SEM analysis was proved. Method of pollen separation from the impaction substrate, polyurethane foam, into homogeneous deposit on Nylon filter was optimized. Representative portion of the deposit was analyzed by SEM. There were taken 485 SEM pictures from 12 samples in 3 localities in the ČR. Pollen grains were identified in 295 SEM and determined into 9 genus and 4 families and grain deformities were quantified to be 24, 18, 50% for Prague, Brezno and Láz localities. Number of pollen increased with total aerosol mass in Prague locality only. Thre aleo identified insect secrction products borochomomes in the samples.
