Abstrakt: [cze][eng] Metylace DNA je jednou z nejčastějších epigenetických modifikací, při které dochází k přenosu metylové skupiny z donorové molekuly S-adenosyl-L-metioninu na 5'pozici cytosinu za vzniku 5-metylcytosinu. Tato reakce je katalyzována DNA metyltransferázami. Epigenetická modifikace hraje důležitou roli v regulaci transkripce, genomovém imprintingu, inaktivaci X-chromozómu a vývoji organismu. Právě její role v regulaci transkripce je důležitá u nádorových onemocnění. Zejména její aberantní formou, hypermetylací, může docházet k umlčení transkripce tumor supresorových genů vedoucí k progresi nádorového onemocnění. This reaction is catalyzed by DNA methyltransferases. Epigenetics modification plays an important role in the regulation of the transcription, genomic imprinting, X-chromosome inactivation and the development of the organism. This role in the regulation of transcription is important for the cancer. Especially the aberrant forms, like hypermethylation, which leads to transcriptional silencing of the tumor suppressing genes leading to the tumor progression, or hypomethylation causing genomic instability.
