PharmDr. Tomáš Šimůnek, Ph.D. Název diplomové práce: Hodnocení cytotoxických účinků vybraných akcelerantů transdermální penetrace u fibroblastů Akceleranty transdermálnej penetrácie uľahčujú prestup liečivej látky cez kožnú bariéru do systémovej cirkulácie alebo do priľahlých tkanív. Oproti klasickým cestám podávania liečiv má tento prístup mnohé výhody, preto v posledných desaťročiach prebieha v tejto oblasti intenzívny výskum. Mechanizmus účinku akcelerantov nie je doteraz presne známy. Reverzibilne znižujú nepriestupnosť kože nešpecifickými interakciami so zložkami stratum corneum (SC), proteínmi a lipidmi, alebo menia rozdeľovaciu rovnováhu medzi SC a liečivom. Cieľom našej práce bolo určenie cytotoxických účinkov vybraných akcelerantov stanovením hodnôt IC50 nových látok syntetizovaných na našom pracovisku s klinicky používanými. V našich pokusoch sme použili myšie embryonálne adherentné fibroblasty 3T3. K stanoveniu viability buniek sme využili MTT test, kedy živé bunky boli schopné redukovať žlté farbivo na modrý formazán. Testy sme realizovali na 96-jamkových doštičkách, kde sme inkubovali bunky s vybranými koncentráciami akcelerantov po dobu 48 hodín. K... PharmDr. Tomáš Šimůnek, Ph.D. Title of diploma thesis: Assessment of the cytotoxic effects of selected transdermal permeation enhancers in fibroblasts Transdermal permeation enhancers facilitate the process of drug penetration trough the skin barrier into the systemic circulation or into surrounding tissues. This approach has many advantages compared to traditional ways of drug administration, hence we observe intense research in this area in last decades. The mechanism of action of enhancers is not exactly elucidated yet. They reduce reversibly barrier properties of skin by nonspecific interactions with components of the stratum corneum (SC), i.e. proteins and lipids, or change the partitioning between the SC and the drug. The aim of our work was to determine the cytotoxic effects of selected transdermal permeation enhancers comparing IC50 values of new substances synthesized in our department with clinically used agents. The mouse embryonic fibroblasts 3T3 cell line was used in our assay. We used the MTT test to determine the viability of cells, only viable cells were able to reduce the yellow dye to blue formazan.
