Souhrn Deprese je systémové onemocnění s komplexní patofyziologií projevující se biologickými, kognitivními a psychopatologickými symptomy. Nalézáme narušení hormonální regulace, imunitní změny a narušení funkce zvláště monoaminergních a indolaminergních neuromediátorových systémů. Farmakologický zásah pomocí antidepresivně působících léků vede k odeznění depresivního syndromu i k úpravě homeostatické nerovnováhy. Jak nervový, tak i imunitní a endokrinní systém využívá k přenosu signálu do vnitra buňky receptory spřažené s heterotrimerickými G proteiny (G proteiny). Předkládaná práce studuje změny vyvolané stresem u experimentálních myší v CNS a na periferii a antidepresivy vyvolané změny na úrovni hladin G proteinů v in vitro C6 gliomové linii a in vivo v tkáních laboratorního potkana. Imobilizační stres u myší vyvolává významné změny v hladinách G proteinových podjednotek ve slezině i v CNS s obdobným profilem odpovědi. Výsledky poukazují na důležitost stresové dopaminergní komponenty v regulaci leukocytárních funkcí a vliv dopaminergní regulace na lymfocytární funkce bez vlivu stresu. Nálezy na gliomové linii pak ukazují specifické antidepresivy vyvolané změny hladin G proteinových podjednotkových profilů a demonstrují tak mechanismus účinku nezávislý na inhibici vychytávání neurotransmiterů, který se... Depression is a complex systemic disorder with exhibiting biological, cognitive and psychopathological symptoms. There are changes in hormonal regulation, immune changes and disturbance in function of especially monoamine and indoleamine neurotransmitter circuits in the CNS. Pharmacologic intervention with antidepressants is treating depressive syndrome as well as homeostatic imbalance. These neurotransmitter systems use in the signal transduction from membrane into the cell receptors coupled with heterotrimeric G proteins (GTP binding proteins).This thesis studies changes in G protein subunit levels induced by stress and psychotropic drugs in the CNS and immune system of experimental mice and antidepressant induced changes in vitro in the C6 glioma cell line an in vivo in the rat tissue. Immobilization stress induces prominent changes in the G protein subunit levels in the spleen and CNS of experimental mice with analogical profile of response. Results show importance of stress dopaminergic component in the leukocyte function regulation and also demonstrate importance of dopaminergic regulation without stress exposition.
