Modulace synaptického přenosu v zadním rohu míšním hraje klíčovou úlohu v nociceptivní signalizaci. Studie z poslední doby naznačují velký význam míšních presynaptických TRPV1 (transient receptor potential vanilloid) receptorů, které na periferii působí jako integrátory nociceptivních podnětů. Mechanismy jejich aktivace a jejich vliv na modulaci synaptického přenosu ale nejsou dosud zcela známy. Předchozí studie také ukázaly vliv řady mediátorů zánětu a cytokinů na funkci TRPV1 receptorů. Cílem naší studie bylo proto ukázat, jak se mění aktivace presynaptických TRPV1 receptorů v míše po aplikaci endogenního agonisty N-oleoyldopaminu (OLDA) u modelu periferní neuropatie, po inkubaci s cytokinemTNFα a jaký je účinek prekurzoru anandamidu N-acylfosfatidylethanolaminu (NAPE). V experimentech byly snímány miniaturní excitační postsynaptické proudy (mEPSC) na neuronech akutních míšních řezů metodou patch clamp. V první sérii experimentů byla testována citlivost na aplikaci endogenního agonisty OLDA 5 dnů po navození modelu periferní neuropatie. Aplikace již nízké koncentrace OLDA (0,2 μM) výrazně zvýšila frekvenci mEPSC na 250 % kontrolní hodnoty, na rozdíl od snímání u kontrolních experimentů kde je ke zvýšení potřeba vysoké koncentrace OLDA (10 μM). Inkubace řezů s TNFα ve druhé řadě pokusů u stejného... (English) Modulation of synaptic transmission in dorsal horn of spinal cord plays a key role in nociceptive signalling. Recent studies have indicated a great importance of presynaptic TRPV1 receptors (transient receptor potential vanilloid) in spinal cord. These receptors act as molecular integrator of nociceptive stimulation on periphery. The way of their activation and the effect on modulation of the synaptic transmission are not clarified yet. Previous studies demonstrated the influence of many inflammatory mediators and cytokins on TRPV1 receptors. The aim of our research was to show changes in activation of presynaptic TRPV1 receptors in the spinal cord following the application of endogenous agonist N-oleoyl dopamine (OLDA) in a model of peripheral neuropathy, after incubation with cytokine TNFα and to show the effect of precursor of anandamide N-acylphosphatidylethanolamine (NAPE). In our experiments, we have recorded miniature excitatory postsynaptic currents (mEPSC) from neurons of acute spinal cord slices by the patch-clamp method. The first series of experiments tested sensitivity to application of the endogenous agonist OLDA 5 days after evoking peripheral neuropathy.
