Primordiální gonocyty (PGC) u některých živočichů, včetně ryb vznikají po oplození v extragonádálním místě z maternálně děděné zárodečné cytoplazmy a během embryogeneze migrují do oblasti budoucích gonád, kde diferencují do gamet. Tvorba a migrační vzory PGC byly studovány u několika modelových druhů zvířat a je známo, že tyto vzory od sebe liší. Jeseteři patří do třídy paprskoploutvých ryb (Actinoptergii), u které je fylogenetická pozice jeseterů mimo skupinu kostnatých ryb a navíc se vývojový vzor jeseterů podobá více obojživelníkům než kostnatým rybám. Touto studií jsme vyvinuli injekační techniku pro vizualizaci PGC jeseterů pomocí GFP nos1 3'UTR mRNA. U jesetera malého (A. ruthenus) jsme zjistili, že jsou PGC specifikovány ve vegetativním pólu embrya. Následně na to jsme popsali migrační trasu PGC. Vzniklé PGC aktivně migrovaly na žloutkově buněčnou hmotu, žloutkovou rozšířeninu a pak byly pasivně přesunuty do gonadální brázdy. Tato studie je důkazem o tom, že jsou PGC specifikovány maternálně děděnou zárodečnou plazmou, nacházející se ve vegetativní části embrya. Způsob specifikace PGC u jeseterů byl podobný žabám, ale migrační vzorec se podobal kostnatým rybám. Dále se nám pro další potřebné studie povedlo úspěšně izolovat PGC. Primordial gonocytes (PGCs) in some animals, including fish arise after fertilization in extragonadal region from maternally inherited germline cytoplasm, and migrate to the future gonads region during embryogenesis, where differentiate into gametes. PGCs formation and migration patterns have been studied in several species models, and it is known that these patterns differ from each other. Sturgeons belong to class ray-finned fishes(Actinoptergii), in which the sturgeon phylogenetic position is an out-group to teleost fishes, the sturgeon development pattern is more similar to amphibians than teleost fishes. In this study, we demonstrate an injection technique for sturgeon PGCs visualization by GFP nos1 3'UTR mRNA. We found that the Sterlet(A. ruthenus) PGCs are specified in the vegetative pole of the embryo. Subsequently, we reported the PGCs migration route. The arisen PGCs actively migrated on the yolky cell mass, yolky extension, and after that passively moved to gonadal ridge. This study provides evidence that the PGCsare specified by maternally inherited germplasm, located in the vegetative part of the embryo. Sturgeon PGCs specification was similar to that of anuras, but the migration pattern resembled that of teleost. Furthermore, we successfully isolated PGCs to next needed studies.
