The work captures the procedure for determining the current state of lean manufacturing and design solutions to increase productivity and fluidity of material flows while adhering to the principles of lean manufacturing.

Construction Project for Station of Fire and Rescue Services in Jaroměř

The objective of this diploma thesis is the processing of the construction technology design as a part of the preparation and implementation of the fire station in Jaroměř. It is a new construction of the main building, which is supplemented by other objects and engineering buildings. The thesis deals with complex design of the implementation of the construction and then focuses in more detail on the installation of the supporting frame structure, which consists of reinforced concrete prefabricated elements. The whole project also includes a technical report on the construction technology project, a study of the implementation of the main technological stages of the main building, solutions for transport routes for construction supply, construction site design, machine set design, technological regulations with assembly diagrams, control and test plan, time plans and balance of resource needs, itemized budget and occupational safety and health plan. The diploma thesis is supplemented by a specialized part dealing with the reconstruction of the area road with a bituminous cover and the implementation of other paved areas within the area of the fire station.

This very important safety element is used during assembly of mainpole high voltage and protect the construction against the robbery of single parts as well as the stability disturbance in the large.
Production of steel structures of halls

Bachelor thesis deals with the production of steel constructions of halls in a complex way. The introductory chapter introduces the advantages and disadvantages of steel constructions, their basic distribution, individual types of constructions and their parts. The following chapters describe the individual phase production of steel constructions of halls, from initial design and negotiations with the investor, through preparation of production, production itself, to final assembly. In the production process are described possible variants of the methods used. Description of the assembly procedure is complemented by the most common cover elements of the hall. The practical part describes the production and installation of a specific steel construction in a selected production plant. The part of the bachelor´s thesis is also the issue of budgeting with indication the individual items that affect the final price of the contract.
Turbine drive for charger unit of bus

Diploma thesis deals with use of combustion chamber to drive the electric bus charging unit. Based on the research and analysis of operation economy, a turboexpander with an air pressure tank is selected to drive the charging unit. A thermodynamic design is created for this variant. Based on this design a unit layout is proposed. Layout drawings are created for the proposed layout.
Subunit c of mammalian F1Fo ATP synthase - from molecular mechanisms of assembly to potential therapies

Mammalian F1Fo ATP synthase plays a crucial role in ATP production through the process of oxidative phosphorylation. Assembly of this multisubunit protein complex requires specific assembly factors. Notably, the assembling of subunit c in mammals requires several factors, yet despite decades of research, the import of subunit c, its incorporation into the inner mitochondrial membrane, and the assembly of monomers towards octameric c-ring remain enigmatic. To shed new light on this process, we first screened for interacting partners of subunit c using a mass spectrometry-based approach. Our screen identified three proteins as the most prominent interactors of subunit c- TMEM70, TMEM242, and c15orf61, which we subsequently characterized. For the initial characterization of the TMEM242 function, we generated both knockdown and knockout HEK293 models. TMEM242 knockdown led to impaired biogenesis and decreased levels of assembled ATP synthase without affecting the content of other OXPHOS complexes. On the contrary, complete deficiency of TMEM242 lead also to the downregulation of complexes I and IV, which indicates that the primary target of TMEM242 is ATP synthase.
