The diagnostic and prognostic ability of selected serum and urinary markers of prostate cancer Abstract Serum prostate specific antigen (PSA) is the only widely approved marker in prostate cancer (PC) diagnosis and follow up after treatment. Its role has remained controversial due to lack of specificity and the risk of overdiagnosis of insignificant PC. The aim of this work was to explore promising markers of PC and to improve current patient stratification to adjuvant treatment. Three main studies were performed using different media (urine and serum). The first study included the evaluation of Engrailed-2 (EN2) - a urinary marker of interest - in 90 patients with localized PC, 30 healthy controls, and 40 patients indicated for prostate biopsy. The second study evaluated 205 men with high-risk PC-features who underwent radical prostatectomy (RP) and were subject to a strict follow-up protocol of ultrasensitive PSA (UPSA) at close time intervals. The ability of particular measurements to predict biochemical recurrence (BCR) and thus the need for adjuvant therapy was assessed using the area under the curve (AUC) and a stratification model was created. Molecular biology of renal tumours in clinical use MUDr. Ivan Trávníček Summary Aim: To identify tissue biomarker that are predictive of the therapeutic effect of sunitinib in treatment of metastatic clear cell renal cell carcinoma (mCRCC). Material and methods: Our study included 39 patients with mCRCC; these were selected from 119 patients who received sunitinib in our hospital between the years 2005 - 2012 according to inclusion criteria of the study. Patients were stratified into two groups based on their response to sunitinib treatment; non-responders (progression), and responders (stable disease, regression). The effect of treatment was measured by comparing imaging studies performed before the initiation of treatment, with those done between 3rd and 7th months of treatment. Histological samples of tumour tissue and healthy renal parenchyma, acquired during surgery of the primary tumour, were examined with immunohistochemistry to detect tissue biomarkers (mTOR, p53, VEGF, HIF1, HIF2, CAIX). The average levels of biomarker expression in both, tumour tissue, as well as in healthy renal parenchyma were compared between the two groups of patients. Title: The effect of material demands and socio-economic status of families on the choice of sport Objectives: Find out if material support in the form of renting hockey gear enables families to overcome the barrier of financial demands when choosing ice hockey as a sport for children. Using the material support may be related to level of socioeconomic status. Therefore, the dependence of using material support and level of the socioeconomic status will be examined. Results can be utilised for better planning of projects which aim at supporting and expanding the membership base of ice hockey or other sports. Methods: In the thesis secondary data analysis method will be used. Moreover, primary data will be collected, using quantitative method of questioning. Results: Failure to provide material support is not a significant barrier for choosing ice hockey as a sport for children. The possibility of material support rather allows parents to easily and quickly provide their kids with hockey equipment in one place. Thanks to this, their children can try ice hockey for the first time. Possibility of renting equipment free of charge means that parents do not primarily look for a reduction of costs on purchasing of hockey gear. Detection of circulating tumour cells and their clinical application in patients with bioptically proven prostate cancer.

1 ABSTRACT Introduction and aim of the study Circulating tumor cells (CTCs) are a promising tool of identifying patients with castration- resistant prostate cancer (CRPC) who will benefit from often demanding cytotoxic therapy. The aim of this work was to evaluate the prognostic significance of CTC in docetaxel-treated CRPC patients. During the project, we also tested the various methods of CTC cultivation and studied their genetic profile as well as the genetic profile of histological specimen at the time of diagnosis. Patients and methods A total of 39 patients who met the CRPC criteria and were indicated for docetaxel chemotherapy were included in the prospective study. Blood collection for CTC analysis was done in all patients before chemotherapy and on the first day of the fourth or fifth cycle of docetaxel. In parallel, CTCs were cultivated. Isolation and detection of CTC was done using the AdnaTest system, which consists of immunomagnetic separation and subsequent detection of mRNA from the CTC lysate. Title: Funding Opportunitites For Ice Hockey Club Slavoj Zbraslav Objectives: The key objective of this thesis is to find out new possibilities of funding the ice hockey club Slavoj Zbraslav, especially from public funds. In addition, proposals for the specific grants, that the ice hockey club can use, are made as a part of the thesis. Methods: The thesis use a qualitative method of an interview to collect the primary data. Further the secondary data are analysed with the methods of descriptive analysis and analysis of the documents. Results: The ice hockey club Slavoj Zbraslav does not use all possible grants for funding. Therefore, some of the funding program were selected and new proposals for grants were suggested.
