5 Abstract Zuzana Skořepová Alteration of transport proteins expression during obstructive cholestasis in rats II Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Pharmacy Background: The aim of the diploma thesis was the verification of cholestatic liver impairment induced by bile duct obstruction lasting for 28 days in rats, by analysis of mRNA and protein expression of liver efflux transport proteins (Bsep, Bcrp, Mrp2, Mdr1, Mdr2, Mrp3, Mrp4). Methods: Wistar rats (n = 6, in each group; weighing 280 to 320 g) were divided into two groups: control rats were sham-operated (Sham) and BDO group (bile duct obstruction lasting for 28 days). mRNA and protein expression changes of the transporters were perfomed by qRT-PCR and Western blot. Results: As compared to Sham group, BDO group showed increase of mRNA expression of Mdr2 transporter to 303%, Mdr1b to 340% and Mrp3 to 9900%. Mrp2 mRNA expression was decreased to 42%. Obstructive cholestasis led to significant increase of protein levels - Bcrp to 160%, Mdr1 to 1010%, Mrp3 to 940% and Mrp4 to 140%. Mrp2, Bsep and Mdr2 protein levels were increased to 31%, 59% and 82%, respectively. Conclusion: Obstructive cholestasis led to decrease of canalicular liver transporters expression and increase of basolateral liver transporters...
Evaluation of the properties of granules and tablets prepared from starch

Zuzana Skořepová Title of Thesis: Evaluation of the properties of granules and tablets prepared from starch In this thesis, the flow properties of granules prepared from potato and/or corn starch respectively, using two types of disintegrants (croscarmellose or sodium starch glycolate) by fluid bed granulation were studied. The particle size distribution, the bulk and tapped density and flow rate of granules and their porosity by gas pycnometer were determined. After the addition of magnesium stearate (0,5 %) the tablets were compressed and their strength and disintegration time were evaluated. The significant effect of type of starch and disintegrants for the disintegration time was proved (p ≤ 0,01); the strength of tablets was significantly (p ≤ 0,01) influenced by only type of disintegrant. The best bulk properties and the shortest disintegration time were observed for the granules made from potato starch in combination with croscarmellose.

The painter Friedrich/ Bedřich Feigl and his family, or The life between nations and metropolises

The presented diploma thesis is concerned with the personality of Friedrich/ Bedřich Feigl, a painter from the classical modern era, and attempts to understand the milieu in which he lived during the two periods of his life. Based on extant sources (letters, memoirs, press of the period, the painter?s own texts) and using historical-antropological optics, Bedřich Feigl, a Prague German of Jewish origin, is placed into a web of social relationships which formed in the first half of the twentieth century. The thesis is divided into two basic parts. The first one focuses on the Prague period, when Feigl, together with his contemporaries, started to build his own artist?s story. The second part deals with Feigl?s life in Berlin, with the possibilities to succeed as a part-foreigner in a foreign country, and also touches Feigl?s perception of his own identity.
